Purpose:Based on the theory of "protein cascade theory",observe the effect of Aβ and the levels of cholesterol which treated with sulforaphane(SFN)on Alzheimer’s disease(AD)rats。To study the protective effects of sulforaphane on Alzheimer’s disease(AD)and its molecular mechanisms in rats。Methods:SPF rats were randomly divided into 7 groups:blank group,sham operated group,Alzheimer’s disease(AD)model group,medicine therapy group,high dose SFN group,medial dose SFN group and low dose SFN group.Alzheimer’s disease(AD)model in rats were copyed by tereotactic the brain’s hippocampus to injection with Aβ1-40(10μg/rat),sham group,saline injection.On the seventh day,each groups were gived drug deliv-ery.Sulforaphane group was given the corresponding concentration,medicine therapy group treated with Piracetam;sham operated group and models group were dealt with NS.Morris water maze was used to test ethology of rats;path-ological changes in hippocampus were observed with HE staining under light microscope;observe the Aβin hippocampus with immunohistochemical;DC Protein Assaysystem combined with COD-PAP to detect the intracerebral level of cholesterol.Results:1.The cellular structure of hippocampus in models group is abnormal,mla-ignmently,the number of neure was decreased significantly,inflammatory cells proliferation;the express of Aβ in hippocampus is rised and increased the levels of cholesterol.2.Comparing the medicine therapy group and models group,the arrangement of structure of hippocampus in medicine group is relatively ordered,the neuron count is restored clearly,the level of Aβexpression and cholesterol were decreased.3.In te SFN control group,te cellular structure of hippocampus almost recovered to normal,the neuron count was increased,the level of Aβexpression and cholesterol were significantly improved.Conclusion:1.SFN can improve the learning and cognitive ability of AD model rats,and enhance the ability of spatial orientation and memory.2.SFN can effectively improve the hippocampus cell morphology and the number of neurons of AD rats,to maintain the overall structure of hippocampus,has neuro-protective effects.3.SFN can reduce the levels of cholesterol in the brain,restrain the express of Aβ,protect the normal physiological function of neurons.SFN can be used for AD patients with a potential prospect of development.
