Objective Explore the postoperative cognitive dysfunction in aging mice and oxidative stress and inflammation reaction and will adjust the relationship between HPA axis to improve the therapeutic effect of cognitive dysfunction in aging mice.Methods Fifty four male mice(3 months old)were divided into young control group(group YC),Two hundred and sixteen male ICR mice(10 months old)were randomly divided into old group(group OC,n=54,as control)and aging model group(n=162),the second one was divided into model group(group OM,n=54);electroacupuncture group(group EA,n=54)and drug group(group OD).According to the execution time after the POCD model established,the model group was divided into model 1 day group(group OMld,n=18),model 3 days group(group OM3d,n=18)and model 7 days group(group OM7d,n=18);the electroacupuncture group was divided into electro-acupuncture 1 day group(group EAld,n=18),electroacupuncture 3 days group(group EA3d,n=18)and electroacupuncture 7 days group(group EA7d,n=18).Mice in group YC received intraperitoneal injection of 0.9%saline(40mg/kg/d,total 5 days)to compare with other groups.Group OM and group EA were adopted the model of partial hepatectomy;group EA received electro-acupuncture,once a day,group OM received the same conditions grab fixed with group EA,this five groups got Openfield and Water Morris maze for detecting cognitive function after electro-acupuncture intervene is over and got execution the second day;group YC and group OC directly got Open-field and Water Morris maze after intervene.Experiment indicators:(1)Detected cognitive function by Open-field.(2)Measured the levels of CRH,ACTH,CORT,MDA,ROS,MDA in cortex,the CRH,ACTH,CORT,MDA,ROS,MDA,SOD in hippocampus and ROS,MDA,SOD in blood serum by enzyme linked immunoadsorbent assay(ELISA).Results(1)Compared with group YC,the central zone time decreased significantly(P<0.05)in group OC.Compared with group OC,the central zone time decreased significantly(P<0.01)in group OM and group EA.Compared with group OM,the central zone time increased significantly(P<0.05)in group EA.(2)Compared with group YC,the latency to the target avoidance increased significantly(P<0.01)in group OC.Compared with group OC,the latency to the target avoidance increased significantly(P<0.01)in group OM1d,group OM3d,group OM7d and group EA.Compared with group OM,the latency to the target avoidance avoidance increased significantly(P<0.01)in group EA3d and group EA7d.(3)Compared with group OC,the levels of SOD decreased significantly(P<0.01)in group OM and group EA.Compared with group OM,the levels of CRH,ACTH,CORT,MDA,ROS decreased significantly(P<0.01 or P<0.05)in group EA.(4)Compared with group YC,the Expression of CRH,ACTH,CORT,MDA,ROS,MDA was increased significantly(P<0.01 or P<0.05)in group OC.Compared with group OM,the Expression of CRH,ACTH,CORT,MDA,ROS,MDA was decreased significantly(P<0.01 or P<0.05)in group EA.Compared with group YC,the Expression of CRH,ACTH,CORT,MDA,ROS,MDA was increased significantly(P<0.01)in group OC.Compared with group OC,the Expression of CRH,ACTH,CORT,MDA,ROS was increased significantly(P<0.01)in group OM.Compared with group OM,the Expression of CRH,ACTH,CORT,MDA,ROS,MDA was decreased significantly(P<0.01 or P<0.05)in group EA.Compared with group YC,the Expression of CRH,ACTH,CORT,MDA,ROS,MDA was decreased significantly(P<0.01)in group EA.Compared with group OC,the Expression of CRH,ACTH,CORT,MDA,ROS,MDA was increased significantly(P<0.01 or P<0.05)in group OM and group EA.Compared with group OM,the Expression of CRH,ACTH,CORT,ROS,MDA was decreased significantly(P<0.01 or P<0.05)in group EA.(5)Compared with group OC,the Expression of SOD was decreased significantly(P<0.01 or P<0.05)in group OM,OD,EA.Compared with group OM,the Expression of SOD was increased significantly(P<0.01 or P<0.05)in group OD,EA.Conclusion Electroacupuncture can reduce the level of oxidative stress in POCD mice,and enhance the activity of antioxidant enzymes,changing the oxidation state of brain,then improve the POCD.
