Objective constructing a new method for sorting basal cell and luminal cell of prostate cancer based on the fluorescent reporter gene,providing a approach for analysis of cell origin of prostat cancer and related research.Methods Fabricating fluorescent reporter genes which contain the objectve promoter.Lentivirus transfecting LNCa P cell line,and sorting the transfected cell by FACS.Westernblot and Q-PCR verifies the sorted cells marker.Results 1.Lentivirus successfully transfect LNCa P cell line。basal and luminal cells are sorted trumphantly 2.result of westernblot and Q-PCR is according with the expectation.Conclusions It is feasible to using fluorescent reporter gene and FACS sorting to get basal cells and luminal cells of LNCa P cell lines of prostate cancer.
