Dibenzocyclooctadiene lignans as the important bioactive components in Schisandra chinensis have been studied for decades.However,fingerprint as the main method for quality control standard of Schisandra chinensis can not combine chemical constituents with bioactivities.In this study,the purpose is to explore the effective relationship between chemical constituents and hepatoprotective of crude dibenzocyclooctadiene lignans from Schisandra chinensis.And it is hoped to predict the bioactive compounds and the best proportion which will contribute to the development of liver-protective drugs.Meanwhile,the technique of extraction and enrichment of dibenzocyclooctadiene lignans were optimized and the dibenzocyclooctadiene lignans have been isolated and identified by different chromatography and spectrum technique.So the following 6 parts will be showed in this thesis.In the introduction,the research progress,objective and significance about this experiment were intruduced.The experiments are divided into 5 parts to introduce.firstly,the extraction and enrichment technique was optimized.The best technique parameter showed that 90% ethanol was used for reflux extraction for 2 times,4 h each time,and solid-liquid ratio of 7:1;the ethyl acetate was used for extract for 5 times;30% ethonal of 6 times and 95% ethonal of 20 times column volume successively were used to obtain crude dibenzocyclooctadiene lignans.Secondly,12 dibenzocyclooctadiene lignans were purified with variety of chromatography techniques.Thirdly,fingerprint of dibenzocyclooctadiene lignans was established,and 17 common peaks were identified in the fingerprint.Forthly,the hepatoprotective effects of 11 batches of crude dibenzocyclooctadiene lignans were evaluated by MTT method.Lastly,spectrum-effect relationship of HPLC specific fingerprint and hepatoprotective effect was analyzed with SPSS 20.0 analysis software.The results showed that schisantherin,schisandrin A and schisandrin B played an important role in promoting liver-protective effect.The extraction,enrichment,seperation,anaysis and bioactivity of dibenzocyclooctadiene lignan were studied which provided the experimental support and new research strategy for the research and development of fructus schizandrae on hepatoprotective effect as drugs and health care products.
