| ObjectiveTo investigate the effect of fatty acid binding protein-5(FABP-5)gene silencing combined with radiotherapy on human esophageal cancer cell line EC9706.MethodsThe interfering SiRNA sequence was designed and synthesized according to FABP-5 gene coding sequence and transfected into EC9706 cells.The experiment was divided into four groups: Control group(Control),FABP-5 silencing group(FABP-5-SiRNA),radiotherapy group(RT),FABP-5 silencing group+radiotherapy group(FABP-5-SiRNA+RT).Immunohistochemical staining was used to detect the expression of FABP-5 in human esophageal cancer tissues,the growth inhibition rate was detected by MTT,flow cytometry was used to detect the apoptotic rate,and Western blot was used to detect the expression of FABP-5 and Caspase-3.ResultsFABP-5 was highly expressed in human esophageal cancer.Compared with Control group,the inhibition rate,apoptosis rate and the expression of Caspase-3 protein in FABP-5-SiRNA group,RT group and FABP-5-SiRNA+RT group increased in turn,and the inhibition of FABP-5-SiRNA+RT group was the most obvious(P<0.05).ConclusionFABP-5 gene silencing and radiotherapy can synergistically inhibit the growth of human esophageal cancer cell line EC9706 and its mechanism is related to the promotion of apoptosis... |
