Objective To observe the effect of Porphyromonas gingivalis on Eph B4-ephrin B2 signal in MC3T3-E1 cells.Methods MC3T3-E1 cells and P.gingivalis were cultured in vitro.The experiment was divided into four groups: one group was treated with rh BMP-2 to induce MC3T3-E1 cells,and the other three groups were co-cultured with P.gingivalis with different MOI values(0,100,200)for 24 hours.Western-blot,RT-PCR and other methods were used to detect the expression of Eph B4,ephrin B2,Runx2,ALP and OPN.Results1.P.gingivalis can cause apoptosis of MC3T3-E1 cells.2.Under the action of P.gingivalis,the expression of Eph B4,ALP and OPN in the experimental group increased with the increase of MOI between Pg and MC3T3-E1 cells.The expression of ephrin B2 and Runx2 decreased at first and then increased.As a positive control,the expression of proteins in the rh BMP-2 group was also significantly higher than that in the negative control group.The difference was statistically significant(P <0.05).Conclusions P.gingivalis may affect bone balance by affecting the expression of Eph B4-ephrin B2 signal in MC3T3-E1 cells.
