Effect Of Traditional Korean Medicine LuRongDaBu Decoction On Nasal Mucosal Inflammation With Allergic Rhinitis Mice

Objective:To explore the effect and mechanism of traditional Korean medicine LuRongDaBu decoction on nasal mucosal inflammation with allergic rhinitis mice.Methods:It randomly divided fifty female BALB/c mice(clean grade,CL)into five groups:no-treament control group,allergic rhinitis model group,LuRongDaBu decoction low dose group,LuRongDaBu decoction high dose group,dexamethasone positive control group.After ten days of practical feeding,except for tno-treament control group other four groups of mice were intraperitoneally injected with a mixture of OVA and alum(0.2mL/injection,5 times).The mouse model of allergic rhinitis was induced by 20%ovalbumin(OVA).In 20 days,except no-treament control group,all the other groups used 20%OVA nasal drops combined with atomization to establish the mouse model of allergic rhinitis,and scored with reference to the mouse behavior score table.After the successful preparation of the model,the three groups of treatment groups were given intragastric drug intervention for 14 days,while the no-treament control group was given intragastric administration of 0.3%normal saline and scored with reference to the mouse behavior score table;all mice were killed one day after the last drug intervention,and their abdominal aortic blood,nasal tissue and lung tissue were taken,and the corresponding methods were used to deal with the samples.The concentrations of cytokines such as IL-4,IL-5,IL-13 and IFN-y in serum of abdominal aorta of mice were detected by ELISA,the nasal tissue sections of mice were stained with HE,the pathological changes of nasal mucosa were observed and evaluated under electron microscope,and the expression of NF-κBp65 protein in nasal mucosa of experimental mice was detected by immunohistochemical method.Western blotting was used to observe the expression of NF-κBp65 and p-p38MAPK protein in mouse lung tissue.Results:1.The results of behavior score of mice showed that compared with the allergic rhinitis model group,the behaviors of scratching nose and sneezing were significantly decreased after oral administration of pilose antler LuRongDabu decoction and dexamethasone solution(P<0.05).2.The results of ELISA showed that compare with the no-treamen control group the level of IL-4,IL-5 and IL-13 in serum of abdominal aorta was significantly higher in allergic rhinitis model group,while the content of IFN-y was significantly decreased in allergic rhinitis model group.Compared with the allergic rhinitis model group,the level of IL-4,IL-5 and IL-13 in abdominal aorta serum of the treatment groups were decreased significantly.After drug intervention that the level of IFN-γ count increased significantly.3.The results of HE staining showed that compared with the no-treamen control group,the model group’s the pathological changes of nasal mucosa was obvious and the complete nasal mucosa was seriously damaged.With a large number of epithelial exfoliation and lymphocyte proliferation that severe edema and hyperemia of nasal mucosa and a large number of eosinophil infiltration in the submucosa(P<0.05).Compared with the results of the model group,the pathological changes of nasal mucosa of mice in each group were improved to some extent,the nasal mucosa was more intact and the infiltration of inflammatory cells and mucosal hyperemia were improved(P<0.05).4.The results of immunohistochemistry showed that compared with the no-treamen control group,in the allergic rhinitis model group,there was a large amount of NF-κBp65 positive expression in the nasal mucosa.The epithelial cells,glandular cells and inflammatory cells were brown-yellow(P<0.05).5.Westernblot gray analysis showed that compared with the blank control group,the positive expression of NF-κBp65 and p-p38MAPK protein in the lung tissue of the allergic rhinitis model group increased significantly,compared with the model group,the positive expression of NF-κBp65 and p-p38MAPK protein in the lung tissue was effectively inhibited by intragastric administration of LuRongDaBu decoction low and high dose groups and dexamethasone solution(P<0.05).Conclusion:1.Traditional Korean medicine LuRongDaBu can inhibit nasal mucosal inflammation in allergic rhinitis model mice;2.The mechanism of action may be related to inhibition of NF-κBp65 and p-p38MAPK positive expression and regulation of Thl/Th2 immune balance.
