Rheum alexandrae Batal.is a plant of the genus Rheum in the family Polygonum,the name of Tibetan medicine is Qumazi;Qumazi in the Si Bu Yi Dian,Jing Zhu Ben Cao and other records,with the effect of clearing heat,detumescence,removing irritability,treating Huangshui disease and malignant ascites.However,currently there is no standard for collecting R.alexandrae.,and there are few studies on its chemical compounds,so it is urgent to clarify its active components and conduct quality control studies.New concept of quality markers(Q-marker),refers to the stability of the Chinese medicinal materials and products,with the function of the traditional Chinese medicine is closely related to the chemical composition,this label material to reflect the effectiveness and safety of Chinese medicine,and quality control,to improve the quality of Chinese medicine and national medicine science evaluation standard to expand the new way of thinking.Objectives1.Textual research on the base plants of Tibetan medicine Qumazi.2.Potential quality markers in R.alexandrae.were analyzed.3.The quality control of Rheum alexandrae.was studied and the draft quality standard was established.Methods1.By consulting the medical records of herbal medicine in the past dynasties and combining with the relevant modern literature,the name,primordium,medicinal site,sexual taste and efficacy of Tibetan medicine Qumazi were investigated.2.Chemical components of R.alexandrae.were analyzed by UPLC-Q-TOF-MS,and mass spectrometry was performed according to the chromatographic retention,accurate molecular weight,characteristic fragment ions and other information of the compounds,so as to determine the chemical attribution and provide reference for the screening of quality markers.3.Screened the identified components by network pharmacology technology,correlated the active components with the target,conducted GO biological function and KEGG pathway enrichment analysis to find the potential active components,and used molecular docking verification to predict the quality markers of R.alexandrae..4.Based on the predicted quality markers,the characters,identification,inspection,extract and content determination of R.alexandrae.in 16 batches of bracts were studied according to the detection method of 2015 edition of Chinese pharmacopoeia(part Ⅳ).Results1.After research and field investigation,the authenticity of Qumazi is recorded as Rheum nobile Hook.f.et.Thoms.,Rheum pumilum Maxim.and Polygonum sibiricum Laxm.,R.alexandrae.is mainly used in the Ganzi Prefecture of Sichuan.2.Identified 45 compounds from R.alexandrae,including tannins and anthraquinones,which initially revealed the chemical constituents of R.alexandrae.,which was used as a reference for the screening of quality markers.3.Through the analysis of network pharmacology,there are 16 potential active ingredient on the 127 targets give play to the role of anti-inflammatory,chrysophanol-8-O-β-D-glucoside,emodin-8-O-β-D-glucoside,emodin,acetylemodin methyl ether glucoside,can be connected with more targets,and molecular docking results show that these ingredients combined with target connection also has good activity,therefore may be give play to the role of anti-inflammatory potential active ingredients,It also plays an anti-inflammatory role by regulating the PI3k-Akt signaling pathway,VEGF signaling pathway,HIF-1 signaling pathway,MAPK signaling pathway,TNF signaling pathway and other signaling pathways.4.The characters and microscopic identification of R.alexandrae.were determined and the TLC method was established.establish the corresponding limit requirements(moisture content 10.87 % or less,the total ash content 9.50 % or less,acid insoluble ash content0.57 % or less,alcohol soluble extract 28.73 % or higher),established its materials of total anthraquinone content determination method and content limits(calculated on dry goods,emodin,chrysophanol,physcion shall not be less than 0.031 %).ConclusionIn this paper,by combining UPLC-Q-TOF-MS and network pharmacology,preliminary bracts big potential,the active ingredient,a study on quality control and R.alexandrae.,implements the quality markers found,provides the reference for the further research and development of Qumazi.
