| Adhatoda vasica Nees.((?))belongs to Adhatoda in Acanthaceae,It is widely distributed in Yunnan,Sichuan and other places in China.The Tibetan medicine classics Crystal Pearl Materia Medica records that Adhatoda vasica Nees.tastes bitter and cold,has the effect of clearing blood fever,liver fever,and chiba fever.It is mainly used for the treatment of rheumatoid arthritis.Adhatoda vasica Nees.,as a commonly used medicinal material in Tibetan medicine,is included in the 2005 edition of the Standard of traditional Chinese Medicine of Yunnan Province.The current quality standard only stipulates the items of character identification,thin layer identification(control medicine),water and extract.It is unable to control quality effectively.The quality markers of traditional Chinese medicine are chemical substances inherent in traditional Chinese medicine and traditional Chinese medicine products or formed in the process of processing and preparation,which are closely related to the functional properties of traditional Chinese medicine.Therefore,So Adhatoda vasica Nees.,as the research object in this study,following traditional medicine experience,according to the quality of traditional Chinese medicine(TCM)markers nature-effect-material principles of theoretical research,the quality of Adhatoda vasica Nees.is found medicine markers,and carries on the quality control research,It will provide a scientific basis for efficacy and safety of the clinical application.Objectives1.Predict potential markers related to medicinal properties and efficacy in the Tibetan medicine Adhatoda vasica Nees,combined with chemical composition studies,find quality markers;2.Improving the quality control method of Tibetan medicine Adhatoda vasica Nees..Methods1.Using molecular docking technology to predict the markers of bitter taste of Tibetan medicine Adhatoda vasica Nees.;2.Using the method of network pharmacology to predict the markers of Tibetan medicine Adhatoda vasica Nees.in the treatment of rheumatoid arthritis;3.Useing the Griess method to screen the anti-inflammatory active parts of the Tibetan medicine Adhatoda vasica Nees.,and use phytochemical separation methods and modern spectrum identification techniques to study its chemical composition;4.Based on the above test results,quality markers were selected,and the quality control of Tibetan medicine Adhatoda vasica Nees.was conducted according to the detection method of the 2015 edition of Chinese Pharmacopoeia(Part four).Results1.There are mainly six subtypes of alkaloids and flavonoids regulating bitter receptors in Adhatoda vasica Nees.,among which vasicine and 5-methoxyvasicine have relatively strong regulatory effects on bitter receptors.Therefore,vasicine and5-methoxyvasicine can be used as markers for the bitterness of Adhatoda vasica Nees..2.Alkaloids and flavonoids in Adhatoda vasica Nees.such as vasicine,N-demethyladhatodine,kaempferol-3-O-rutinoside,quercetin-3-O-sophorosidemay be the pharmacological basis of Adhatoda vasica Nees.in the treatment of rheumatoid arthritis.3.The dichloromethane extraction site of the Tibetan medicine Adhatoda vasica Nees.has the strongest anti-inflammatory activity,and 14 components were isolated and identified as: vasicinone(1),vasicine(2),7-methoxyvasicine(3),7-hydroxyvascinone(4),7-methoxyvascinone(5),syringaresinol(6),iatrointelignan A(7),jatrointelignan B(8),acernikol(9),3-indolcarbaldehyde(10),vanillin(11),1′,2′-dehydromarmesin(12),ethyl 4-hydroxybenzoate(13),ethyl4-hydroxyphenylacetate(14).It has been reported in the literature that compounds 1,2,3,6,10 have anti-inflammatory activity,so they can be used as a reference for qualitymarkers.4.Based on the above results,it is clear that vasicine is the quality marker of Adhatoda vasica Nees..The TLC identification method was determined,and the content of anemone in 11 batches of anemone was determined and the corresponding limit was prescribed(in terms of dry products,anemone in anemone is≥0.46 %).The fingerprint was established(similarity≥0.900).The characters and microscopic characteristics of Adhatoda vasica Nees.were determined.The water content,total ash content,acid insoluble ash content and extract content of 11 batches of Tibetan medicine anemones were determined and the corresponding limits were established(water content≤12.3 %,total ash content≤19.5 %,acid insoluble ash content≤0.08 %,water-soluble extract content≥21.2 %).Conclusions1.Revealed the Tibetan medicine Adhatoda vasica Nees.and its medicinal properties and efficacy-related markers,and identified vasicine as a quality marker.2.Improve the quality control method of the Tibetan medicine Adhatoda vasica Nees.,which provides a scientific basis for the establishment of quality standards and the effectiveness and safety of clinical application. |
