| Objective To observe the protection from the Rho kinase inhibitor after ICH and the impact on the expression of eNOS.Methods One hundred and forty rats were randomly divided into 5 groups,including sham-operated group,ICH 1d group,ICH 3d group,Fasudil 1d group and fasudil 3d group.Their brains were taken out at planned time.The tissue around the hematoma was taken,the brain water content was measured by dry-wet weight method,the apoptosis was observed by TUNEL fluorescence,while the level of IL-6 and TNF-α in brain tissue was detected by ELISA kit.Changes in expression of ROCK2 and eNOS were examined by Western blot.Results1.The brain tissue water contents of rats in ICH 1d and 3d groups were significantly higher than that in the sham operation group.After fasudil treatment,the water contents of brain tissue at 1d and 3d were significantly lower than that of ICH 1d and3 d groups.(P<0.05)2.The apoptosis rates of rats in ICH 1d and 3d groups were significantly higher than that in the sham operation group.After fasudil treatment,the apoptosis rates of the perihematomal tissue at 1d and 3d were significantly lower than that of ICH 1d and3 d groups.(P<0.05)3.IL-6 and TNF-α relative contents of rats in ICH 1d and 3d groups were significantly higher than that in the sham operation group.After fasudil treatment,IL-6 and TNF-α relative contents of the perihematomal tissue at 1d and 3d were significantly lower than those of ICH 1d and 3d groups.(P<0.05)4.ROCK2 and eNOS mRNA levels of rats in ICH 1d or 3d groups were significantly higher than that in the sham operation group.After fasudil treatment,ROCK2 and eNOS mRNA levels of the perihematomal tissue at 1d and 3d were significantly lower than those of ICH 1d and 3d groups.(P<0.05)5.ROCK2 and eNOS protein levels of rats in ICH 1d and 3d groups were significantly higher than that in the sham operation group.After fasudil treatment,ROCK2 and eNOS mRNA levels of the perihematomal tissue at 1d and 3d were significantly lower than those of ICH 1d and 3d groups.(P<0.05)Conclusion The Rho kinase inhibitor,fasudil,can alleviate brain edema,inflammatory response and neuronal apoptosis in rats with cerebral hemorrhage,and its mechanism may be related to eNOS.It plays a protective role and provides new therapeutic direction for the treatment of cerebral hemorrhage. |
