| Background Gastric cancer has the characteristics of high morbidity,high mortality,easy transfer,etc.The molecular mechanism underlying the development of gastric cancer is still unclear.Long non-coding RNAs(lncRNAs)are a class of strands,with a length greater than 200 nucleotides,which are not capable of encoding a protein or can encode only micropeptides.In recent years,its role in the development of tumors has gradually been recognized.Based on our previous work,we will investigate the role of lncRNA FMO6 P in the progression of gastric cancer.Therefore,we could improve the understanding of the regulation mechanism of gastric cancer progression.MethodsWe used qRT-PCR to detect the expression of lncRNA FMO6 P in 100 gastric cancer mucosa tissues and adjacent normal mucosa tissues.The expression of lncRNA FMO6 P in gastric cancer cell lines MGC803 and HGC27 was up-regulated by CRISPR/Cas9 activation.The proliferation,migration and invasion abilities of gastric cancer cell lines MGC803 and HGC27 overexpressing lncRNA FMO6 P were detected by transwell assay and CCK8,respectively.The level of EMT markers,β-Catenin,AKT and AKT phosphorylation in gastric cancer cells was detected by Western Blot.Gastric cancer cells MGC803 and HGC27 overexpressing lncRNA FMO6 P and control groups were injected into the subcutaneous or peritoneal cavity of nude mice respectively to study the effect of lncRNA FMO6 P on the subcutaneous tumor formation and peritoneal dissemination of gastric cancer cell lines.ResultsThe expression of lncRNA FMO6P was down-regulated in mucosal tissues of gastric cancer,and it is significantly correlated with tumor size.lncRNA FMO6 P could inhibit the proliferation,invasion and migration of gastric cancer cell lines MGC803 and HGC27 and the EMT process.At the same time,lncRNA FMO6 P could prohibit the subcutaneous tumor formation and peritoneal dissemination of gastric cancer cell lines in nude mice.LncRNA FMO6 P overexpression reduced β-catenin in gastric cancer cell lines MGC803 and HGC27,and although it could not regulate the AKT,it inhibited the phosphorylation of serine at position 473 of AKT.ConclusionsLncRNA FMO6P is lowly expressed in gastric cancer tissues and is significantly associated with tumor size.lncRNA FMO6 P inhibits the invasion,migration and EMT of gastric cancer cells by decreasing β-catenin and inhibiting AKT phosphorylation. |
