Cinnamon recorded in Chinese pharmacopoeia is the lauraceae plants Cinnamomum cassia Presl dry bark.Clinical studies have shown that cinnamon has anti-tumor,anti-inflammation,anti-microbico,anti-oxidation,hypoglycemic and lipidlowering effects.Cassia twig is the lauraceae plants Cinnamomum cassia Presl tender twig,which is often used for cold,edema,palpitations and so on.Cinnamon recorded in European pharmacopoeia and British pharmacopoeia is the Cinnamomum Cinnamomum verum J.Presl dry bark.It is widely used in traditional medicine treatment of respiratory system,digestive system and gynecological diseases.The powders of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark are similar in appearance,which is easily confused and difficult to identify.But the clinical use and chemical basis of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark have great differences.In order to identify the similarities and differences among Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark and ensure the safety,effectiveness and quality control of the clinical use of the medicinal materials,we constructed the LC-MS and GC-MS chromatographic system for the chemical composition characterization to establish the related chemical composition data information database and clarify the overall quality control of the material basis aimed at Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark in the project.On this basis,an features exclusive qualitative identification method based on fingerprint was established to reveal the differences components of the three.Furthermore,the method of simultaneous quantitative analysis of multiple components was constructed based on single standard for determination of multiple components,and the data were analyzed and mined by combining with chemometric analysis.Finally,the overall quality control system was constructed,which can be used for the quality evaluation of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark,the traditional medicine in cinnamomum.1.Establish a Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark chemical composition data information database.The volatile oil of Cinnamomum cassia bark and Cinnamomum verum bark was determined by GC-MS,and the chemical structure was studied by NIST mass spectrometry database retrieval and manual analysis.148 chromatographic peaks were detected from Cinnamomum cassia bark essential oil and 77 components were identified,accounting for 95.77% of the total chromatographic peak area.121 chromatographic peaks were detected from Cinnamomum verum bark essential oil and 73 compounds were identified,accounting for 96.37% of the total chromatographic peak area.There were 32 common components in Cinnamomum cassia bark essential oil and Cinnamomum verum bark essential oil,accounting for 83.12% and 85.61% of the total area of essential oil respectively.A total of 24 chemical components including 3 flavanols,8 alkaloids,3simple aromatic compounds,8 simple phenylpropanoids and 2 fatty acid compounds were identified from Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark by LC-MS.There were 15 common components in Cinnamomum cassia bark and Cinnamomum verum bark.The chemical constituents of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark were characterized to clarify the material basis of the overall quality control of the three.2.Established an features exclusive qualitative identification method.The fingerprint of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark was established by UPLC,and the similarity evaluation was used to ensure the stability and reliability of the medicinal materials.There were 7 common peaks in the fingerprint of Cinnamomum cassia bark and Cassia twig respectively,and 10 common peaks in the fingerprint of Cinnamomum verum bark.Ten components were identified by the chemical components of fingerprinting,including procyanidin B2,norboldine,cinnamtannin B1,coumarin,2-hydroxycinnamaldehyde,cinnamyl alcohol,cinnamic acid,cinnamaldehyde,2’-methoxycinnamaldehyde and eugenol.Cinnamtannin B1 and eugenol were the unique components of Cinnamomum verum bark.Then,chemometric analysis were used for comprehensive analysis of the fingerprint of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark to quickly and effectively distinguish and revealed the difference components of the three.The main difference components in order of VIP size were as follows: cinnamaldehyde > cinnamic acid >2’-methoxycinnamaldehyde > cinnamtannin B1> coumarin,which laid a foundation for the further determination of the indicator components of the content determination.3.Establish the method of simultaneous multi-components content determination.Based on the results of fingerprints combined with chemometrics analysis,nine differential and directive components were selected to determine the content of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark.This new method named single standard for determination of multiple components(SSDMC)used procyanidin B2 as the internal control of proanthocyanidins and cinnamic acid as the internal control of phenylpropanoids to determinate of multiple components for Cinnamomum cassia bark,Cinnamomum verum bark and Cassia twig simultaneously.Respectively using this method and external standard method calculated the content of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark,and the results of SSDMC / ES showed that there was no significant difference between the two methods,indicating that this SSDMC method was feasible to determine of the three.The content results of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark were analyzed by chemometric analysis to quickly and effectively distinguish the three.This method can be considered an effective and reliable method for distinguish of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark.In conclusion,the chemical composition of Cinnamomum cassia bark,Cassia twig and Cinnamomum verum bark was characterized to established the related chemical composition data database.On this basis,this study built the quality control analysis strategy of "one method for multiple evaluations" based on LC-MS and GC-MS analysis for non-volatile components and volatile components,and established the features exclusive qualitative identification method based on fingerprint and the simultaneous multi-components content determination based on single standard for determination of multiple components.Then,the data were analyzed and mined by combining with chemometric analysis,and we formulated relevant standards to provide demonstration research for establishing a unified quality control system for similar drugs.
