Objective:With the rapid development of the world economy,people are no longer satisfied with food and clothing,but begin to pay more and more attention to physical,mental,environmental protection and other issues and impacts,among which physical diseases are the most noticeable.Cancer has become a global leading cause of death,the serious influence and harm to human body and health,Although the social economy and scientific and medical technology have made rapid progress in recent years,various types of drugs and diagnostic tests and treatments for diseases have been developed rapidly,but does not effectively reduce the mortality,which caused by the cancer related treatment,and also the side effects of drugs.So it’s important to find new drugs that are less toxic or even have no side effects.Stahlianthus involucratus is a common Chinese herbal medicine,which has been used for thousands of years in China.It has been widely used in promoting blood circulation and dispersing blood stasis,reducing swelling and relieving pain,treating and improving injury caused by falls,rheumatic bone pain and snake and insect bites.The drug has been scientifically proven to have a good anti-tumor effect,and its chemical composition can kill cancer cells.This experiment intends to use the method of separation,purification and pharmacological experiment to study the effect of Stahlianthus involucratus toxicology and elucidate its effect.Methods: 1.Isolation and purification of total extracts of Stahlianthus involucratus1.1 Various extraction methods of Stahlianthus involucratus were explored to explore the effects of different extraction methods and factors on the real extraction rate of Stahlianthus involucratus.1.2 The total extracts of Stahlianthus involucratus were separated and purified with macroporous adsorption resin,and different elution sites could be obtained.The anti-tumor activity of various eluent sites of Stahlianthus involucratusin tumor-bearing mice was evaluated.The elution sites with the best activity were selected by silica gel column chromatography and then analyzed and purified by silica gel column chromatography.2.Study on the toxic mechanism of the active site of Stahlianthus involucratus2.1 Acute toxicology experiment SPF grade BALB/c mice were gavaged at different concentrations.By comparing with the control group,the characteristics of body weight,hair and spirit were observed,the death,organ index and biochemical indexes were recorded,and half of the deaths were calculated.2.2 Subacute toxicology experiment Kunming mice were treated by long-term gavage with different drug concentrations.By comparing with the control group,the characteristics of body weight,hair and spirit were observed,and death,organ index and biochemical indexes were recorded.Results : 1.Isolation and purification of total extracts of Tiantian Notoginseng1.1 Different extraction methods were tested to obtain the extraction results,and the results were compared.It was concluded that the extraction rate was the highest when Stahlianthus involucratus was extracted by 70%ethanol heated reflux method.1.2 Anti-tumor activity experiments showed that the site eluted with 70%ethanol had the best anti-tumor effect,followed by the site eluted with 95%ethanol.2.Experimental Study on the Toxicity of 70% Ethanol Elution Fraction of Panax tuber2.1 In the acute toxicological experiment,the body weight,mental state,number of deaths,organ index and biochemical indexes of mice were observed to confirm that Stahlianthus involucratus could indeed cause influence and injury to the body under a certain dose.2.2 In the subacute toxicological experiment,the body weight,mental state,number of deaths,organ index and biochemical indexes of mice were observed to confirm that a small amount of Stahlianthus involucratushad no damage to the body organs in a chronic manner.Conclusion : Under the premise of the anti-tumor mechanism of Stahlianthus involucratus,the dosage range of Stahlianthus involucratus should be determined to prove its safety and reliability,and its dosage should be clarified to provide scientific basis and safety guidance for the promotion of clinical application of Stahlianthus involucratus.
