OBJECTIVE: Study the effect on the invasion and migration of human nasopharyngeal carcinoma cell line(CNE-2)from volatile oils in Stahlianthus involucratus.METHODS: The inhibitory effect on the CNE-2 cell proliferation of volatile oils was observed by Cell Counting Kit8(CCK8)test in vitro.The inhibitory effect on the CNE-2 cell invasion and migration of volatile oils was observed by Wound-healing assay and Transwell assay.The expression levels of MMP-2,MMP-9 and VEGF were detected by Western blot.The expression of gene was analyzed by RT-PCR.RESULTS:CNE-2 cell proliferation was resulted in significant inhibition(P<0.01)and in a dose and time dependent manner and IC50 for them were 70.296?g·ml-1,63.749?g·ml-1,59.532?g·ml-1,respectively,after volatile oils were incubated with CNE-2 cell for 24,48 and 72 h.Rate of CNE-2 cell invasion and migration was reduced and accompanied in a concentration-dependent manner by treated with volatile oils for 48 h.CONCLUSIONS: Expression levels of MMP-2,MMP-9 and VEGF were all down-regulated in a concentration-dependent manner.Volatile oils can inhibit CNE-2 cell invasion and migration with down-regulation of MMP-2,MMP-9 and VEGF expression. |
