| Objective: To study the histological changes of pedicled buccal fat pad in repairing buccal mucosa defect. Methods: Defects of the same size(1cm*1cm)were prepared in bilateral buccal mucosa of white rabbits in New Zealand. One side was repaired with buccal fat pad, while the other side was not treated.Photographic records were used to observe the healing at 1st,2nd and 4th week after operation. The wound area was calculated by Image J software and statistically processed by SPSS23.Paraffin sections were made for HE staining,IHC staining and Masson staining, and then observed under an optical microscope. Results: The healing time of the wound surface repaired by buccal fat pad was shorter and the mucosa epithelialization was obvious. The results of HE staining showed that at the 1st,2nd and 4th week after operation, compared with the experimental group, the epithelialization of the wound area took a long time and more inflammatory cells infiltrated in the natural healing group. The results of immunohistochemical staining showed that there was a difference in the degree of epithelialization between the experimental group and the control group at the first week, and the two groups were completely epithelialized at the 4th week.Through Masson staining,it was found that the amount of collagen increased gradually with the extension of time, and the difference was significant at he first week and second week. Conclusion:The healing time of buccal mucosa defects repaired by pedicled buccal fat pad was short, the epithelialization was obvious, the inflammation was less, and the wound healing was better. |
