| Objective:To investigate the expression levels of Treg cells and cytokines TGF-β1,IFN-γ,IL-4 and IL-17 A in peripheral blood of patients with myelodysplastic syndrome(MDS)and acute myeloid leukemia(AML),and to explore the differences in immune status of patients with MDS and AML at different stages and their clinical significance.Methods: 1.A total of 20 clinically newly diagnosed and untreated MDS patients were collected and divided into relatively low-risk group(10 cases)and relatively high-risk group(10 cases)according to the risk of IPSS-R,15 AML patients,and 10 healthy subjects were selected as normal control group.2.Flow cytometry was used to detect the expression ratio of Treg cells in peripheral blood.3.The expression levels of cytokines TGF-β1,IFN-γ,IL-4 and IL-17 A in serum of peripheral blood were detected by ELISA.Results:(1).The expression proportion of Treg cells in MDS group and AML group was significantly higher than that in control group(P < 0.05),and the expression proportion showed an increasing trend with the progression of the disease(P < 0.05).Compared with the relatively low-risk group,the expression ratio of Treg cells in the relatively high-risk group and AML group was significantly increased(P <0.05).Compared with the high-risk group,the expression ratio of Treg cells in AML group was increased(P < 0.05).(2)The expression levels of TGF-β1,IFN-γ,IL-4 and IL-17 A in MDS group were higher than those in control group(P < 0.05),The expression levels of TGF-β1 and IL-4 in AML group were higher than those in control group(P < 0.05),and there was no significant difference in the other two cytokines;Compared with the relatively low risk group,the expression levels of TGF-β1 and IL-4 in the relatively high risk group and AML group were significantly increased(P < 0.05),while the expression levels of IFN-γ and IL-17 A in the relatively high risk group and AML group were significantly decreased(P < 0.05).Compared with the relatively high risk group,the expression level of TGF-β1 in AML group was increased(P < 0.05),and the differences in IFN-γ,IL-4 and IL-17 A were not statistically significant compared with the relatively high risk group.Conclusion: The immune status of patients with MDS and AML at different stages is different,and immune abnormalities may be involved in the occurrence and development of MDS. |
