| Background:Seborrheic dermatitis is a common chronic inflammatory skin disease in clinical practice,recurrent,may be accompanied by varying degrees of itching.Its exact etiology and pathogenesis have not been fully elucidated,and current studies have shown that it may be related to genetic factors,immunodeficiency,fungal infections,hormone levels,neurological factors and environmental factors,and Malassezia is a common pathogen causing seborrheic dermatitis.Antifungal therapy is a common treatment for seborrheic dermatitis of the head.With the massive use and abuse of antibiotics,more and more resistant strains appear,but the available antifungal drugs are limited,so it is necessary to develop new drugs that are not resistant and have few side effects.Antimicrobial peptides have great development prospects and application value in many fields such as medicine,food,and aquaculture,especially in the medical field.The main component of Medi Hair Spray Dressing is a new polymer complex,and its core component is antimicrobial peptides,which is a microbial source.It has antibacterial,oil control,repairing skin lesions,changing the local microbial growth environment,playing a role in remissin and treatment for seborrheic dermatitis,with good clinical application prospect.In this paper,the antibacterial activity and effectiveness of Medi Hair Spray Dressing were preliminarily discussed through the experimental study of the antibacterial activity of Medi Hair Spray Dressing against Malassezia in vitro,providing a theoretical basis for the clinical treatment of seborrheic dermatitis of the head.Objective:Two in vitro drug sensitivity test methods,Ager plate diffusion method(punch method)and Microliquid-Based dilution method,were used to observe the in vitro antibacterial activity of Medi hair spray dressing and its main components(Microbial antimicrobial peptide)against common Malassezia on the surface of seborrheic dermatitis lesions of the head.Methods:1.For Medi Hair Spray Dressing,microbial antimicrobial peptide was used as the experimental group,and Caile lotion(concentration 2%ketoconazole lotion),Blank solvent of Medi Hair Spray Dressing was used as the control group,and the diameter of the inhibition zone was compared in each group.Dilute the stock solution of Medi Hair Spray Dressing by agar plate diffusion method(punch method),and the concentrations are 0.8mg/g、0.2mg/g、0.04mg/g、0.008mg/g、0.0016mg/g、0.00032mg/g.Microbial antimicrobial peptide stock solutions were diluted and the concentrations were 0.8mg/g 、 0.2mg/g 、 0.04mg/g 、0.008mg/g、0.0016mg/g、0.00032mg/g.The Caile lotion stock solutions were diluted and the concentrations were 20mg/g、5mg/g、1mg/g、0.2mg/g、0.04mg/g、0.008mg/g.Blank solvent of Medi Hair Spray Dressing stock solutions were diluted and the concentrations were 562.5 mg/g、140.625mg/g、28.125 mg/g、5.625 mg/g、1.125 mg/g、0.225 mg/g。The positive control group without liquid medicine was set up,while the negative control group was set without liquid medicine.After incubation at 37℃for 72 h in the temperature incubator,the diameter d(mm)of its inhibition zone was measured using a digital vernier caliper.2.For Medi Hair Spray Dressing,microbial antimicrobial peptide was used as the experimental group,and Caile lotion(concentration 2%ketoconazole lotion),Blank solvent of Medi Hair Spray Dressing was used as the control group,and the MIC(minimum inhibitory concentration)values of each group were compared.With the microliquid-based dilution method,glyceryl monooleate was used as liquid media for susceptibility testing according to the NCCLS-M27 A protocol in the yeast dilution method established by the National Committee for Clinical Laboratory Standards,the inhibitory effect of Medi Hair Spray Dressing diluted to different concentrations and its main components(microbial antimicrobial peptide)on Malassezia in vitro was determined on a 96-well microplate,and compared with that of Caile lotion and Blank solvent of Medi Hair Spray Dressing diluted to different concentrations.the positive control group was inoculated with bacteria and no liquid was placed,and the negative group was not inoculated with bacteria.The MIC(minimum inhibitory concentration)values of Medi Hair Spray Dressing,microbial antimicrobial peptide,Caile lotion and Blank solvent of Medi Hair Spray Dressing were determined.Results:1.Ager plate diffusion method(punch method): 0.8mg/g、0.2mg/g、0.04mg/g、 0.008mg/g 、 0.0016mg/g 、 0.00032mg/g concentrations of compound Medi Hair Spray Dressing for bacteriostasis diameter of Malassezia are 31.87 mm 、30.11 mm 、21.47 mm 、19.89mm、14.53mm、0mm.0.8mg/g 、 0.2mg/g 、 0.04mg/g 、 0.008mg/g 、 0.0016mg/g 、0.00032mg/g concentrations of compound Microbial antimicrobial peptide for bacteriostasis diameter of Malassezia are 24.59mm、20.10mm、16.17mm、0mm、0mm、0mm.20mg/g、5mg/g、1mg/g、0.2mg/g、0.04mg/g、0.008mg/g concentrations of compound Caile Lotion for bacteriostasis diameter of Malassezia are 32.07mm、30.49mm、25.53mm、22.42mm、17.38mm、0mm.562.5 mg/g、140.625 mg/g、28.125 mg/g、5.625 mg/g、1.125 mg/g、0.225 mg/g concentrations of compound Blank solvent of Medi Hair Spray Dressing for bacteriostasis diameter of Malassezia are13.55mm、0mm 、0mm 、0mm 、0mm 、0mm.2.Microliquid-Based dilution method: Medi hair spray dressing and Microbial antimicrobial peptide had varying degrees of antibacterial effect against Malassezia.Medi hair spray dressing MIC is0.977ul/ml;Microbial antimicrobial peptide MIC is 3.907 ul/ml.Conclusions:1.Medi Hair Spray Dressing and Microbial antimicrobial peptide inhibited Malassezia to varying degrees in vitro,and its antibacterial effect was positively correlated with drug concentration.2.Determination of MIC of Medi Hair Spray Dressing by Microliquid-Based dilution method is 0.977ul/ml;Microbial antimicrobial peptide MIC is 3.907 ul/ml;3.Medi Hair Spray Dressing is safe,not easy to produce drug resistance,few side effects,inhibit Malassezia easy to use,and can be used for the treatment of seborrheic dermatitis of the head in clinical practice. |
