Ectodermal-neural Cortex 1 Affects The Biological Function Of Lung Cancer Through The MAPK Pathway | | Posted on:2022-07-15 | Degree:Master | Type:Thesis | | Country:China | Candidate:C W Wu | Full Text:PDF | | GTID:2504306335451554 | Subject:Internal Medicine | | Abstract/Summary: | PDF Full Text Request | | Backgroud and Objective: Ectodermal-neural Cortex 1(ENC1)high expressed in lung cancer tissues was screened out from the TCGA database.The objective of this study was to investigate the effects of ENC1 on the biological functions of lung cancer cells.Methods: The expression of ENC1 was investigated by q PCR between 28 lung cancer and para-cancer tissues and the relationship between its expression and clinicopathological features was evaluated.The expressions of the normal lung cell line and lung carcinoma cell lines were estimated by q PCR and Western blotting.Then we knockdowned the expression of ENC1 in A549 and H1299 cells by transfecting si ENC1 and verified the transfection efficiency by q PCR and Western blotting.After transfecting si ENC1,we estimated the proliferation and invasion of cells by CCK-8 and transwell assay.Meanwhile,some proteins related to migration and invasion were estimated by Western blotting.Plasmid was used to upregulate the expression of ENC1 in lung cancer cell lines A549 and H1299,and related functional tests were conducted to verify the effects of ENC1 on lung cancer.At last,the subcutaneous xenotransplantation tumor model was established in nude mice to study the effects of ENC1 downregulation on tumor growth in lung cancer cells.Results: The expression of ENC1 in lung cancer tissues and lung carcinoma cells was significantly higher than that in adjacent tissues and the normal lung cell.Cell proliferation,migration and invasion abilities in lung carcinoma cell lines transfected with si ENC1 were decreased significantly compared with the control group.Further experiments showed that knockdown of ENC1 significantly down-regulated the expressions of MMP2,MMP9,N-cadherin and up-regulated the expression of E-cadherin.Besides,ENC1 downregulation reduced the expressions of p-JNK,p-ERK and p-P38.After overexpression of ENC1,the proliferation,migration and invasion of oe-ENC1 transfected cells were decreased compared with the control group.Finally,downregulation of ENC1 could inhibit the growth of tumors by analyzing of nude mouse model of lung cancer.Conclusion: Above all,ENC1 is involved in the proliferation,migration and invasion of lung cancer,and may be an effective blocking target and a new breakthrough in the treatment of lung cancer. | | Keywords/Search Tags: | ENC1, non-small cell lung cancer, migration, invasion, proliferation, MAPK | PDF Full Text Request | Related items |
