Objective: nephrotic syndrome is a common glomerular disease.there are many pathological types of nephrotic syndrome,so every pathological type has different prognosis.In this study,we selected SD rats to establish adriamycin nephrotic syndrome model,then some rats were given berberine by gavage treatment.we compared with adriamycin-induced nephropathy in rat in the 24-hour urinary protein,plasma protein,blood lipid,renal function and renal histomorphology to evaluate whether berberine hydrochloride can delay the progress of the nephrotic syndrome.At the same time,we made a research on what exactly the mechanism that the berberine hydrochloride apply in adriamycin nephrotic syndrome.Methods: First,we need to establish adriamycin nephrotic syndrome model,so we selected 40 healthy male SD rats and they were randomly divided into normal group(Normal),model group(Model),berberine treatment group(BBR)and prednisone treatment group(PED).Meanwhile,30 rats were injected with adriamycin 5mg/kg through tail vein.Then we collected the 24-hours urine in the first and the second week after modeling.While succeeding modeling,we continue feeding these rats about 8 weeks,then rats in Berberine group were given intragastric administration of 100mg/(kg.d),and prednisone group were given intragastric administration of 1mg/(kg.d).After 8 weeks,we killed four groups of rats,then collected their fasting blood and kidney tissue.We used the automatic biochemical analyzer to analyse liver function,renal function,24-hour urine protein,blood lipid and blood glucose,PAS staining was used to observe the pathological changes of kidney tissue in the four groups,and the expression of NF-κB was tested by immunohistochemistry.Results: For the model group,the 24-hour urinary protein quantity after 2 weeks was higher than that of the normal group,the difference was statistically significant(P<0.05);the 24-hour urinary protein quantity and renal function in the berberine group and prednisone group were lower than those in the model group,the difference was statistically significant(P<0.05).In the model group,the glomerulus was enlarged,mesangial area was widened,glycogen deposition was seen in renal tubules,and the nuclei of renal tubular epithelial cells fell off and become voids;the pathological changes in Berberine group and prednisone group were less than those in the model group.Comparing with model group,berberine and prednisone groups could regulate dyslipidemia,and the difference was statistically significant(P<0.05).Comparing with prednisone,berberine has the advantage of reducing blood glucose.Comparing with the model group,the expression levels of NF-κB and IL6 in Berberine group and prednisone group were lower,and the expression level of SOD in serum increased(P < 0.05).Conclusion: Berberine can decrease the level of urinary protein in adriamycin nephropathy rats,reduce the renal injury;the mechanism is that berberine may reduce the expression levels of NF-κB and IL6,increasing the level of serum SOD,inhibiting inflammatory response,inhibiting oxidative stress,and then play the role of renal protection.
