| Bamboo juice is one of natural drinks,which is obtained from the stems of Moso Bamboo(Phyllostachys edulis)and other species of the same genus.According to the Chinese medicine books “Qianjin prescription” and “Ancient and modern record and test prescription”,it is commonly used to treat various diseases,such as cough and phlegm.In this study,Moso bamboo juice was extracted with ethyl acetate and n-butanol,and the main compounds were identified by column chromatography and spectral analysis.Based on compounds 1-26 as references,precisely qualitative and quantitative the main contents by UPLC-Q-TOF-MS in fresh bamboo juice and nine kinds of bamboo juice oral liquid.At the same time,the sugar content and additives in nine kinds of bamboo juice oral liquid were also determined by ion exchange chromatography and HPLC.In addition,the compounds isolated from bamboo jucie were screened for activity.The main results are as follows:1.Fresh bamboo juice was separated and purified by HP-20 macroporous resin and LH-20 glucan gel,and 26 compounds were obtained.On the basic physic-chemical constants and spectroscopic analysis such as UV and NMR and MS,26 compounds were identified as: 3-Methoxy-4-hydroxybenzyl alcohol(1),β-hydroxypropiovanillone(2),1-(4-Hydroxy-3-methoxyphenyl)-glycerol(3),3,5-dimethoxy-4-hydroxydihydrocinamaldehyde(4),Syringate(5),Syringic aldehyde(6),p-E-Coumaric acid(7),triandrin(8),Ferulaldehyde(9),Coniferol(10),β-Arbutin(11),tachioside(12),4-Hydroxy-3,5-dimethoxyphenyl-β-D-glucopyranoside(13),3,4’-dihydroxy-propiophenone-3-β-D-glucopyranoside(14),β-hydroxypropiovanillone(15),(+)-balanophonin(16),3-[2,3-Dihydro-2-(4-hydroxy-3,5-dimethoxyphenyl)-3-(hydroxymethyl)-7-methoxy-5-benzofuranyl]-2-propenal(17),2,6-Dimethoxy-p-benzoquinone(18),2-Furaldehyde(19),Adenosine(20),2-hydroxy-5-methoxyphenyl-β-D-Glucopyranoside(21),(+)-lyoniresinol-3α-O-(β-D-Glucopyranosyl)(22),(+)-5’-Methoxyisolariciresinol-3α-O-β-D-glucopyranoside(23),(-)-Lyoniresinol 3α-O-(β-D-Glucopyranoside)(24),7S,8R,8’R-5,5′-dimethoxyariciresinol-4-O-β-D-glucopyranoside(25),Syringaresinol 4’-O-β-glucopyranoside(26).Compound 25 was new compound.Compounds 7,9,11,12,21,and 23 were isolated from fresh bamboo juice for the first time.2.A new method for the detection of fresh bamboo juice compounds was established by UPLC-Q-TOF-MS,and the accuracy,precision and sample recovery rate of the method were verified.Then a qualitative and quantitative analysis was performed on fresh bamboo juice and nine kind of bamboo juice oral liquid.It was found that there are similarities and differences between fresh bamboo juice and nine kinds of bamboo juice oral liquid.The commonality is that there are some identical compound ingredients,the difference is that the content of the compounds is significantly different.Basically,the content of compounds in fresh bamboo juice is much larger than that in bamboo juice oral liquid.In addition,the difference is that there are some different compounds in different bamboo juice oral liquid,and the content is slightly different.3.The sugar content of nine kinds of bamboo juice oral liquid was determined by ion exchange chromatography.The results showed that the main carbohydrate components were found to be arabinose,with maximum contents reaching up to 33230.78 mg/L.Determination of additives in bamboo juice oral liquid by HPLC.And the contents of three additives were 1484.00 to 2754.25 mg/L,230.11 to 471.44 mg/L,and 1797.48 mg/L,respectively.4.Twenty-six compounds isolated from fresh bamboo juice were evaluated for their antiinflammatory activity,XOD inhibitory activity and PTP1 B inhibitory activity,and the results showed that compound 14 exhibited XOD inhibitory activity.All compounds have no antiinflammatory and PTP1 B inhibitory activity. |
