| Objective:The purpose of this study is to compare the effects of ELISA,WB,PCR and other techniques on the pathogenesis of ulcerative colitis and the transformation of JAKs/STATs signal pathway key factors in rats with ulcerative colitis.To analyze the effect of Xiao Zhenqiu prescription Anchang decoction on JAKs/STATs signal pathway in rats with ulcerative colitis,and to explore the characteristics of inflammation and transformation of ulcerative colitis.In order to reveal the possible use of Anchang decoction in the treatment of ulcerative colitis and the clinical application of Anchang decoction,we can rely on the basis of disease recognition and prevention and control of ulcerative colitis at the same time.Methods:(1)Sixty healthy SPF SD rats,half female and half male,weighing180 ~ 220 g.After adaptive feeding,they were randomly divided into blank group,model group,Lizhu Changle group,low,middle and high dose groups of Anchang decoction,with 10 rats in each group.(2)According to the article,except for the blank group,all the rats were modeled with 2,4,6-trinitrobenzenesulfonate(TNBS)/ ethyl alcohol(50%ethanol calculated according to the body weight of 100 mg.kg-1TNBS+ and so on).The rats were intragastrically fed on the second day after the establishment of the model,and the rats were killed and the standard samples were taken after 14 days of administration.(3)Experimental objects: Rats in each group and their abdominal aortic blood and colonic tissue.(4)Experimental method and detection content:During the experimental period,the spirits,defecation,food and other conditions of the rats in each group were observed,and the active index scores of progressive diseases were scored.The pathological analysis was performed in the rat colon group,and the condition of colon mucosal injury in rats was evaluated by the index of colon injury(colon macroscopic damage index,CMDI),and the condition of tissue disease in the rat colon mucous membrane group was evaluated by Geboes index.RT-PCR determination of IL-6,JAK2,STAT3 and SOCS-3m RNA in rat colonic tissue JAK2,p-STAT3,STAT3 and SOCS-3 in rat colonic tissue were detected by;Western blot and the contents of IL-6,TNF-α,IL-17 and IL-10 in serum were detected by;ELIS A.The data obtained from the actual test are shown in the table of mean ±standard deviation(X ±s).SPSS 17.0 soft parts are used for systematic analysis,single factor ANOVA is used for multi-group analysis,and t-test is used for inter-group ratio.The difference was shown in the table of P < 0.05,and the meaning of the difference was shown in the table of P < 0.05.The data obtained are processed and plotted with Graph Pad.Results:(1)The disease symptoms of rats in each group showed obvious disease symptoms in the UC model group,and the DAI score was significantly higher than that in the blank group(P < 0.05).After administration,the symptoms of the rats in each treatment group were improved in different degrees,and the score of DAI in the treatment group was lower than that in the blank group(P < 0.05),and the therapeutic effect was the best in the middle dose group of Anchang decoction.(2)According to CMDI and Geb oes scores,the scores of UC model group were higher than those of blank group,and the scores of each treatment group were lower than those of UC model group(P < 0.05,0.05),and the scores of each treatment group were lower than those of UC model group(P <0.05),and the scores of each treatment group were lower than those of UC model group(P < 0.05).In the middle dose group of Anchang decoction,the recovery of colonic mucous membrane injury was the best.(3)The expression of IL-6,JAK2,STAT3 and SOCS-3m RNA in the colonic tissue of rats in each group changed: compared with the blank group,the contents of IL-6,JAK2,STAT3 and SOCS-3m RNA in the model group were higher than those in the blank group.Compared with the model group the contents of JAK2 STAT3 and IL-6m RNA in each treatment group decreased while the content of SOCS-3m RNA increased significantly(P <0.05).(4)The protein contents of JAK2,p-STAT3,STAT3 and SOCS-3 in the colon group of rats in each group changed: compared with the blank group,the protein contents of JAK2,p-STAT3,STAT3 and SOCS-3 in the model group increased(P < 0.05).Compared with the model group the contents of JAK2 p-STAT3 and STAT3 protein in all treatment groups decreased while the content of SOCS3 protein increased significantly(P < 0.05).(5)The contents of IL-6,TNF-α,IL-17 and IL-10 in serum of rats in each group: compared with the blank group,the contents of IL-6,TNF-α,IL-17 and IL-10 in the model group increased(P < 0.05).Compared with the model group,the contents of T NF-α,IL-17 and IL-6 in all treatment groups decreased and IL-10 increased significantly(P < 0.05).Conclusion(s): Anchang Decoction can significantly improve the inflammatory state of rats with ulcerative colitis.It can reduce the expression levels of serum pro-inflammatory factors IL-6,TNF-α,and IL-17,and increase the expression level of anti-inflammatory factor IL-10.High,while down-regulating the expression of IL-6,JAK2,STAT3 m RNA and its JAK2,STAT3 protein,and up-regulating the expression level of SOCS-3 m RNA and SOCS-3 protein,the mechanism may be through the regulation of IL-6/JAK2/STAT3/SOCS-3 Signal pathways play a role in preventing and treating ulcerative colitis. |
