Object: The research purpose of this subject is to use acupuncture at Jiaji points of the neck to treat blood pressure,nitric oxide(NO),angiotensin 2(Ang2),and noradrenaline in L-NAME-induced hypertensive rats.The regulation of NE,SOD activity,malondialdehyde(MDA),and other indicators,and analyze their mechanism of action.To further clarify the effect of acupuncture at the cervical Jiaji points on hypertensive rats,it is expected to provide new theoretical and experimental data for the study of acupuncture and moxibustion treatment of essential hypertension and its mechanism.Methods: 1.For 40 SD male rats,10 SD rats were randomly identified and classified into the blank group,and the other 30 were classified into the hypertension model group,of which 10 were induced by the L-NAME method.Blood pressure model group,10 hypertension models +western medicine treatment group(captopril 10 mg/kg),10 hypertension models + acupuncture group.After the model was successfully modeled,the rats were treated with western medicine or acupuncture at Dazhui,Shenmen,and Zusanli for a week,and the blood pressure of the rats after model creation and treatment was recorded.2.After the experiment,the rats were anesthetized by intraperitoneal injection of 10% chloral hydrate solution 3 ml/1kg,and blood was quickly taken from the common abdominal aorta of the rats,and the contents of NO,T-SOD and MDA in the serum were determined using the kit;Use enzyme-linked immunosorbent assay(ELISA)to determine the content of NE,ET-1 and Ang2 in serum.3.Immediately after the blood collection,the rats were killed,the heart was removed,the left ventricle was isolated,and the respective weights of the heart and left ventricle were accurately weighed,and the left ventricular mass index(LVMI)was calculated.A part of the left ventricle was cut out and fixed with 10%formalin solution for HE staining.4.Extract RNA from rat heart tissue,and qRT-PCR to detect the mRNA expression levels of TNFα and TGF-β1 in the four groups of myocardial tissues.Result: 1.The blood pressure of the three groups of L-NAME induced hypertension modeling was significantly higher than that of normal blood pressure.Through acupuncture and moxibustion in the large spinal canal,Shenmen,and Zusanli rats,the blood pressure of the rats was significantly reduced,which was close to the level of normal rats,which was significantly different from the untreated group.2.After the L-NAME induced hypertension model,the left ventricular mass index level of rats increased compared with normal rats,and after acupuncture the large spinal canal,Shenmen,Zusanli treatment,the left ventricular mass index of rats The level was suppressed,and it was close to returning to normal.After induction of hypertension by L-name,the myocardial cells in the myocardial tissue of the model group rats are hypertrophy,edema becomes obvious,discrete error,proliferation of fibrous tissue can be seen between the muscle bundles,and local fibrous tissue hyaline changes in the myocardial tissue of the acupuncture group The cardiomyocytes in the medium are slightly mutated,mildly disordered,and mildly hypertrophic and edema.There is a small amount of proliferating fibrous tissue between the muscle bundles,and the infiltration of inflammatory cells is not obvious.The morphology of the myocardial tissue cells has been significantly improved.3.The levels of malondialdehyde(MDA),norepinephrine(NE),ET-1,and serum Ang2(angiotensin)in the serum of hypertensive rats were significantly higher than those of normal rats,but after acupuncture After treatment of Dazhugu,Shenmen,and Zusanli,the levels of malondialdehyde(MDA),norepinephrine(NE),ET-1,and serum Ang2(angiotensin)were suppressed.,The activity of superoxide dismutase(SOD)and the level of nitric oxide(NO)in the serum of rats were significantly lower than those of normal rats.After acupuncture at Dazhui,Shenmen,and Zusanli,the superoxide dismutation Enzyme(SOD)activity and nitric oxide(NO)levels have increased.4.After the L-NAME induced hypertension model,the mRNA expression levels of death factors TNFα and TGF-β1 in rat tissues were higher than that in normal rats.However,after acupuncture treatment,inflammation in rat tissues The mRNA expression levels of factor TNFα and TGF-β1 were reduced.Conclusion:1.Acupuncture at Dazhui,Shenmen,and Zusanli can reduce L-NAME-induced hypertension in rats.2.Acupuncture at Dazhui,Shenmen,and Zusanli can improve the cardiac morphology and myocardial tissue morphology of hypertensive rats induced by L-NAME.3.Acupuncture at Dazhui,Shenmen,and Zusanli can alleviate hypertension in rats by regulating the levels of NE,SOD,NO,MDA,Ang2 and ET-1.4.Acupuncture at Dazhui,Shenmen,and Zusanli can alleviate hypertension in rats by regulating the levels of TNFα and TGF-β1. |
