Objective:Use data mining technology to study the prescription rules of modern doctors for the treatment of knee synovitis,use relevant algorithms to establish possible prescriptions for traditional Chinese medicine for treatment of knee synovitis,and analyze them through network pharmacology,and finally guide clinical medication.Predict the role of related mechanisms.Methods:The clinical observations published in the academic databases of CNKI,VIP,and Wanfang are used as the source of literature to collect medical record information that meets the diagnosis.Analyze the frequency of prescription traditional Chinese medicines,nature,flavour and channel tropism of of herbs,and use SPSS software to carry out correlation and cluster analysis to get the rules of medication and new prescriptions,and discuss them with Chinese and Western medicine.Put the herbs in the prescriptions obtained from the analysis into the TCMSP database and the CNKI database,and use the relevant rules to find the effective ingredients and their targets,and put the obtained targets into the Uniprot database for screening;use the GeneCards database to find the relevant knee synovitis Target;establish a component-target-disease network;use STRING database to construct a PPI network,and perform GO and KEGG enrichment analysis on it to predict the potential mechanism of the prescription for the treatment of knee synovitis.Results:①According to the reading of the original text and the two-person comparison,a total of 117 literatures were included.There were 101 Chinese medicine prescriptions and 152 Chinese medicines.The top ten Chinese medicines used were Angelicae Sinensis Radix,Licorice,Coicis Semen,Achyranthes bidentata Radix,Rhizoma Atractylodis,C yathula officinalis Kuan,Astragali Radix,Atractylodes macrocephala Rhizoma,Poria,Alis matis Rhizoma.The first three meridians are divided into liver,kidney,and spleen meridians.The Four Properties of traditional Chinese medicine are mostly warm,calm and cold,and the Five Tastes are mostly bitter,pungent,and sweet.According to the association rules,it is found that the top five drug pairs are Angelicae Sinensis Radix and Chuanxiong Rhizoma;Rhizoma Atractylodis,Achyranthes bidentata Radix and Coicis Semen;Rhizoma Atractylodis and Phellodendri chinensis Cortex;Coicis Semen and Phellodendri chinensis Cortex;Rhizoma Atractylodis,Phellodendri chinensis Cortex and Coicis Semen.and the top five trustworthy pairs are Persicae Semen and Carthami Flos;Persicae Semen,Car thami Flos and Chuanxiong Rhizoma;Persicae Semen,Angelicae Sinensis Radix and Carthami Flos;Stephaniae tetrandrae Radix,Coicis Semen and Rhizoma Atractylodis;Rhizoma Atractylodis,Phellodendri chinensis Cortex and Achyranthes bidentata Radix.According to the cluster analysis,4 types of prescriptions were obtained,cluster 1:Persicae Semen,Carthami Flos,Rehmanniae Radix,Angelicae Sinensis Radix,Chuanxiong Rhizoma,Paeo niae Radix Rubra,cluster 2:Gentianae Macrophyllae Radix,Angelicae pubescentis Radix,Saposhnikoviae Radix,Notopterygii rhizome Et Radix,Taxilli Herba,Spatholobi Caulis,licorice,Cyathula officinalis Kuan,Astragali Radix,cluster 3:Salviae miltiorrhizae Radix Et Rhizoma,Chaenomelis Fructus,Cinnamomi Ramulus,Polyporus,Atractylodis macroce phale Phizoma,Alismatis,Poria,Radix Paeoniae radix Alba,cluster 4:Stephaniae tetrandr ae Radix,Dioscoreae spongiosae Shizoma Rhizoma Atractylodis,Phellodendri chinensis Cortex,Coicis Semen,Achyranthes bidentata Radix.According to the TCMSP database,a total of 51 active ingredients of the clustering prescriptions were obtained,and a total of 228 standard genes were obtained by screening the corresponding targets of the activeing redients in the Uniprot database.436 targets of knee synovitis were screened in the GeneCards database.The main effective components in the prescription are quercetin,luteolin,kaempferol,baicalein,β-carotene,ellagic acid,β-sitosterol and other chemical components.The protein interaction network established on the STRING platform shows that VEGF A,TNF,IL-6,PTGS2,CXCL8,and MMP9 are the most important target proteins.②Based on the STRING platform,perform GO and KEGG enrichment analysis on 45 common targets of diseases and prescriptions,and found that in the biological process,the top five are the cellular response to lipopolysaccharide,inflammatory response,negative regulation of apoptotic process,positive regulation of transcription from RNA polymerase II promoter,lipopolysaccharide-mediated signaling pathway;in cell composition,the top five are extracellular space,extracellular region,external side of plasma membrane,proteinaceous extracellular matrix and cytosol In the analysis of KEGG pathway,the TNF signaling pathway,TOLL receptor signaling pathway,NF-κB signaling pathway,HIF-1 signaling pat hway,and VEGF signaling pathway are closely related to knee synovitis.Research conclusion:① In the treatment of synovitis of the knee joint,modern physicians used the methods of activating blood to resolve stasis and eliminating dampness while adding tonifying drugs to achieve the effect of both internal and external treatment.The high-frequency drugs are Angelicae Sinensis Radix,Licorice,Coicis Semen,Achyrant hes bidentata Radix,Rhizoma Atractylodis,Cyathula officinalis Kuan,Astragali Radix,Atr actylodes macrocephala Rhizoma,Poria,Alismatis Rhizoma..②The main effective components of clustering prescriptions composed of Persicae Semen,Carthami Flos,Rehmanniae Radix,Angelicae Sinensis Radix,Chuanxiong Rhizoma,Paeoniae Radix Rubra for the treatment of knee synovitis are quercetin,luteolin,kaempferol,baicalein,β-carotene,tannin Flower acid,β-sitosterol and other chemical components,the targets are VEGFA,TNF,I L-6,PTGS2,CXCL8,MMP9,etc.By affecting the TNF signaling pathway,TOLL recept or signaling pathway,NF-κB signaling pathway,HIF-1 signaling pathway,VEGF signaling pathway and other pathways,it can inhibit inflammation,resist angiogenesis,promote a poptosis,and prevent cartilage damage.
