ObjectiveBladder cancer(BCA)is a common malignant tumor of the urinary system,which mainly originates from the urothelium and seriously endangers human health.BCa is characterized by high recurrence and easy metastasis,but the mechanism is still unclear.The origin and anatomical structure of bladder tissues are also different.Studies have shown that the recurrence and metastasis of bladder cancer are closely related to the location(sub organ)of the disease.The purpose of this project is to analyze the sequencing data and clinical information and follow-up information of bladder cancer in online database to preliminarily determine the relationship between the location of bladder cancer and clinicopathological grade and prognosis,epithelial mesenchymal transition(EMT)score,immune cell infiltration and tumor mutation load(TMB),and to construct organ like model,so as to provide theoretical basis for molecular typing of bladder cancer.MethodsUsing online data of bladder cancer expression and gene mutation,chi square test was used to analyze the relationship between the location of bladder cancer and clinicopathological grading and staging.Immune infiltration analysis was performed with R-Pack GSVA at R3.6.T test was used to analyze the distribution of immune cell infiltration in bladder sub organs.The difference was statistically significant(P<0.05).The tissue samples of bladder cancer and normal paracancerous tissues were collected and cultured in organoid medium to construct three-dimensional organoid model.ResultsCombined with the sequencing data that have been submitted to the database,according to the location of the tumor,that is,the bladder triangle,the bladder roof,and the bladder wall(anterior wall,posterior wall,and bilateral wall).It is found that the trigonometric region of the bladder cancer has higher staging,faster disease progression and worse survival rate,which has statistical significance.The mutation load and epithelial mesenchymal transition score of bladder triangle tumor were the lowest,but there was no statistical difference;The infiltration degree of T cells,Tfh cells and Treg cells in the triangle area was the lowest with statistical difference.The organ like model of bladder cancer was successfully constructed,which laid a foundation for molecular typing of bladder cancer.ConclusionsThe location of bladder cancer is related to the clinical stage of the tumor,the prognosis of the patient and the infiltration of immune cells.Meanwhile,the organoid model of bladder cancer was successfully constructed by using organoid culture technology.The successful establishment of this model will explain the relationship between the location of bladder cancer and the occurrence and prognosis of patients,and provide a theoretical basis for the specific molecular biological mechanism,which has a broad application prospect.Figure 8 table 1 reference 56.
