The Analysis Of Famous And Experienced Chinese Medicine Doctors In Treating Esophageal Cancer Based On Data Mining

ObjectiveThis study comprehensively collects the medical cases of famous and experienced Chinese medicine doctors in the treatment of esophageal cancer,building a database based on the Chinese medicine inheritance platform system and uses association rules and system entropy clustering algorithm to explore the rules of prescriptions.At the same time,network pharmacology was used to the study of mechanism of representative drug combinations,so as to provide certain evidence support and reference value for rational drug use in clinical treatment of esophageal cancer.MethodsA comprehensive computer search of the China Network Knowledge Infrastructure(CNKI)database,Viper Journal database(VIP),Wan fang data.Besides,cases from famous and experienced national,provincial and civic Chinese Medicine doctors for esophageal cancer such as contemporary famous old Chinese medicine typical medical case collection、TCM master medical case record series、TCM master pro syndrome essential series and famous old Chinese medicine prescriptions experience series were manually collected.TCMISS(V2.5)was used for data mining.The syndrome principle,herbs association rules and core new formula of these cases were analyzed and summarized.The "Pinellia-Poria"drug pair was selected,and its related components and targets were retrieved based on the Chinese Medicine Systematic Pharmacological Analysis Platform(TCMSP).The targets of esophageal cancer were found based on DisGeNET database,and the network diagram was estabilshed by Cytoscape 3.7.2.GO function and KEGG pathway enrichment analysis based on the potential targets were constructed to explore the mechanism of "Pinellia-Poria" on the treatment of esophageal cancer.ResultsA total of 209 medical cases were collected from these databases and the above books.The results showed that the five common types of patterns were mutual association of phlegm and blood stasis,deficiency of qi and Yin,obstruction of phlegm and qi stasis,and deficiency of fluid and heat stasis.The warm,cold,sweet and bitter herbs were used more often and they were composed of spleen,stomach,lung and liver mostly.The commonly used 10 traditional Chinese medicine were pinellia,poria,licorice,astragalus membranaceus,atractylodes,tangerine peel,hedyotis diffusa,clematis root,fritillaria thunbergii,raddle;The association rules indicated that pinellia,poria,atractylodes,licorice,Astragalus membranaceus,tangerine peel had the strongest correlation.Nine groups of new formulars were obtained:①Inula flower,raddle,walnut kernel,pollen typhae;②Allium macrostemon,sandalwood,loofah,hemp seed,cassia twine;③Scorpion,centipede,mulberry,silkworm;④Scutellaria scutellaria,angelica sinensis,bean root,peach kerne,safflower;⑤Radix rehmanniae,dendrobium officinalis,radix ophiopogonis,polygonum japonicus,radix japonicus;⑥Licorice,green bark,agilawood,dendrobium,radix ophiopogonis;⑦The ammoniac stone,bai ying,huang jing,caulis spatholobi,placenta homin;⑧Ligusticum wallichii,saponin thorn,cinnamon,pangolin,myrrh;⑨Cinnamon,Aconite,Cannon Ginger,Dried Ginger,Asarum.A total of 46 potential targets,including Aktl,TP53,FOS,VEGFA,Jun and MMP9,were obtained from common drugs on Pinellia-Poria,and TNF,PI3K-Akt,MAPK and other signaling pathways were involved in the treatment of esophageal cancer.ConclusionIn general,when treating esophageal cancer,famous and experienced Chinese medicine doctors stress the value of strengthening spleen and stomach;They are skilled in using herbs in activating qi circulation、removing phlegm stasis、supplementing Qi and nourishing Yin.Moreover,they also pay attention to the combination of disease and syndrome."PinelliaPoria Cocoa" combination therapy of esophageal cancer may be achieved through the intervention of TNF,PI3K-Akt,MAPK pathways.
