| Objective:Establish an ultra-high performance liquid chromatography-mass spectrometry(UPLC-MS/MS)method to detect methamphetamine,amphetamine and furosemide in body tissues.Further use this method to detect and understand the effect of furosemide on the changes in the tissue distribution of methamphetamine in mice,,and provide an experimental basis for the identification of diuretic drugs to accelerate drug excretion in criminal cases.Methods:1.Establish a UPLC-MS/MS method for the detection of methamphetamine,amphetamine and furosemide,optimize the pretreatment method^establish a calibration standard curve,and detect the accuracy and precision of intraday and interday.2.Establish a dosing model:male BALB/c mice were reared adaptively for three days and randomly divided into 4 groups(blank control group,furosemide control group,methamphetamine control group,methamphetamine combined furosemide group).Each group is divided into 6 subgroups(0.25 hour,0.5 hour,1 hour,2 hours,4 hours and 8 hours).After intraperitoneal administration according to the animal experiment grouping scheme,the animals were sacrificed at each time point,take whole blood,brain and liver tissues,and use UPLC-MS/MS to detect the content of methamphetamine,amphetamine and furosemide in the whole blood and tissues.Results:1.Establish a UPLC-MS/MS detection method for methamphetamine,amphetamine and furosemide.2.Compared with the methamphetamine group,in the methamphetamine+furosemide group,the concentration of methamphetamine in the blood and brain tissues decreased(P<0.05);compared with the furosemide group,in the methamphetamine+furosemide group,the concentration of furosemide in blood,brain tissue and liver tissue decreased(P<0.05).Conclusion:1.Establish a UPLC-MS/MS method to determine the content of methamphetamine,amphetamine and furosemide in mouse tissues,which is simple to operate and has good recovery,accuracy and precision.It can be applied to the detection of methamphetamine,amphetamine and furosemide in cases.2.The use of furosemide will not change the distribution trend of methamphetamine into the body,but it can affect the distribution concentration of methamphetamine in different tissues;at the same time,methamphetamine will also affect the distribution concentration of furosemide in different tissues.3.The detection of diuretics and drugs in biological samples can provide evidence to support the qualitative nature of the case. |
