In this study, the determination method for Methamphetamine and its metabolism Amphetamine in biological samples has been developed. The distributions in addictive animals have been investigated. The results of this study can provide the reference for further study.To establish the determination method for Methamphetamine and Amphetamine in biological samples, the preparation of the bio-meterial is expressed as follow: Samples are alkalinized to pH14, then extracted by the cyclohexane, after centrifuging, the solvent are evaporated at 70℃and the residue is derivatized with acetic anhydride.Gas chromatography is introduced in this paper to determine Methamphetamine and its metabolite Amphetamine. GC analysis is performed using an coated fused silica capillary column (30 m×0.25 mm i.d. 0.25μm film thickness, USA). The column oven temperature is initially held at 80℃, then programmed to reach 140℃, at a rate increase of 30℃/min, then programmed to reach 170℃, at a rate increase of 3℃/min, then programmed to reach 260℃, at a rate increase of 20℃/min, and held for 1 min. The total run time was 21 min. The temperatures of the injector and the detector are set at 250℃and 300℃, respectively. The carrier gas (nitrogen) flow rate is 1.0 ml/min. 4-phenylbutylamine is used as internal standard.The method proposed is used for determinating Methamphetamine and Amphetamine in plasma, urine, heart, liver, spleen, lung, kidney and brain samples. This method is evaluated by linearity, extractive recovery, analytical recovery, within-day and between-day precision. The method is sensitive, accurate, precise and reproducible, and suitable for clinical diagnosis, forensic identification and further study.The distribution of Methamphetamine and Amphetamine is investigated in 12 addictive rats. Addictive rats mode is set up by administered Methamphetamine solution (10 mg/kg, 4 d), and then killed 2 hours later. Heart, liver, spleen, lung, kidney, brain, plasma and urine samples are collected. The concentrations of Methamphetamine and Amphetamine in tissues are determined with Gas Chromatography. The result shows that Methamphetamine and Amphetamine are distributed broadly in tissues of addictive rats, showing difference levels in these tissues.The determine method for Methamphetamine and its metabolite Amphetamine in biological samples is established in this paper. The distributions of Methamphetamine and Amphetamine in addictive animals are also studied. The results can provide a valuable method and reference for clinical diagnosis, forensic identification and further study.
