| Objective:UroVysion,which is commonly used in fluorescence in situ hybridization(FISH)test for bladder cancer diagnosis has lower sensitivity in the diagnosis of early grade bladder cancer than advanced bladder cancer,and its effectiveness in Chinese patients with bladder cancer has not been fully proved.We hope to find a new FISH probe combination for the diagnosis of bladder cancer patients in China,And preliminary evaluation of its clinical application value.Methods:Chromosome microarray analysis(CMA)was performed on bladder cancer tissue samples(n=15)from our hospital.Based on these data,two chromosomal regions were selected,and a two-color FISH probe combination was designed.Urine samples of 41 patients with bladder cancer were collected for FISH detection,and another 20 patients with urinary malignancies excluded were the control group.After the FISH tests are completed,analyze the FISH data.Compared with the clinical data of patients,the clinical application value of the new probe FISH detection was evaluated preliminarily.Results:1、The chromosomal CNV in 15 cases of bladder cancer was tested by chromosomal microarray analysis,The common chromosomal abnormalities include deletion of 17p,9p,9q and amplification of 1q,5p,12q.The most frequently amplified chromosomal region 1q21.1-1q44 and the most frequently deleted chromosomal region 17p13.2-17p11.2 were selected to design a new FISH probe.2、The overall sensitivity and specificity were 80.5%and 90.0%respectively.According to TNM stage,the sensitivity of Non muscle invasive bladder cancer(Ta,T1)and Muscle invasive bladder cancer(T2-T4)was 78.3%and 83.3%respectively.The sensitivity of low-grade urothelial cancer and high-grade urothelial cancer was 75%and 82.8%respectively.The sensitivity of diagnosis of primary bladder cancer was 80.6%,and the sensitivity of recurrent tumors was 80.0%.Conclusions:1、Chromosomal CNV of 1q21.1-1q44 and 17p13.2-17p 11.2 are common in Chinese bladder cancer patients,which may be a new target for FISH test of bladder cancer.2、The test exhibits good sensitivity and specificity in the diagnosis of bladder cancer.Compared with the UroVysion FISH test,it improved the sensitivity for diagnosis of early bladder cancer.At the same time,only two probes were used in this test,which reduces the cost of test and also reduces the complexity of the experiment,which has potential clinical application value.Moreover,the significant differences in the abnormal cell proportion of high-grade and low-grade tumors suggests the potential value in evaluating the prognosis of patients with bladder cancer.However,the amount of samples collected in this experiment is limited,and it can only preliminary assess its value in the diagnosis of bladder cancer patients in China.In the future,it is necessary to collect more samples and evaluate its clinical application value. |
