Objective:The purpose of this study is to study the expression of Deoxythymidylate kinase(DTYMK)in patients with hepatocellular carcinoma,analyze the correlation between DTYMK expression and clinical parameters,and explore the prognosis of DTYMK for hepatocellular carcinoma and diagnostic value.Methods:1.The DTYMK single gene expression profile of hepatocellular carcinoma was derived from TCGA(The Cancer Genome Atlas)and the database GEO,and Western blot and real-time fluorescent quantitative polymerase chain reaction(RT-qPCR)were performed on 12 pairs of liver cancer and adjacent tissue specimens.To study the protein and mRNA expression levels of DTYMK in liver cancer tissues,and to analyze and compare its expression differences in liver cancer and adjacent tissues.2.Download the clinical data set of hepatocellular carcinoma in the database,analyze the relationship between DTYMK expression and clinical parameters,prognosis and diagnostic value,and screen potential signals that are differentially activated by DTYMK in liver cancer through gene enrichment analysis(GSEA analysis)path.3.We transfected MHCC97H and Hep-3B cells with DTYMK-siRNA and NC-siRNA,and measured the transfection efficiency with the help of Western blot and RT-qPCR test,using CCK-8 test,EdU test,flow cytometry,Transwell and other methods to study the effect of DTYMK on the biological functions of liver cancer cells.Results:1.Western blot,RT-qPCR and bioinformatics analysis results showed that compared with adjacent tissues,DTYMK is highly expressed in liver cancer tissues.2.The expression of DTYMK is related to tumor size and degree of invasion(T stage),clinical stage,histological grade and survival status,but has nothing to do with gender and age.Survival analysis found that high expression of DTYMK is significantly related to the poor survival rate of liver cancer patients.The univariate and multivariate Cox analysis showed that the high expression of DTYMK is an independent risk factor related to the prognosis of liver cancer patients.The expression of DTYMK reached a high value of the area under the curve(AUC)in the receiver operating characteristic curve(ROC),thus effectively distinguishing normal liver tissue from liver cancer tissue.GSEA analysis showed that signaling pathways related to arginine and proline metabolism are related to high expression of DTYMK.3.After silencing DTYMK in vitro,the proliferation,migration and invasion of liver cancer cells were inhibited,indicating that DTYMK may become an oncogene through its role in the cell cycle and DNA replication pathway.Conclusion:The expression level of DTYMK in hepatocellular carcinoma tissues is significantly higher than that in adjacent tissues.The high expression of DTYMK is related to the poor prognosis of liver cancer patients and is an independent risk factor affecting the prognosis of liver cancer.The expression of DTYMK has a higher AUC value in ROC,suggesting that DTYMK has the potential to be a diagnostic biomarker for liver cancer.
