| Objective: We established a mouse model of acute asthma induced by ovalbumin(OVA),and the observed the effect of Chan long Ding Chuan Decoction on the pathological changes of lung tissue and the level of inflammatory factors in each group of mice,and explore the therapeutic effect and possible mechanism of Chan long Ding Chuan Decoction on the mouse model of acute asthma induced by ova,so as to provide a new theoretical basis for the selection of innovative drugs for the clinical treatment of acute asthma.Methods: 60 ICR mice were randomly divided into five groups,12 in each group.They were normal group,model group,dexamethasone group(2mg/kg),Chan long Ding Chuan Tang Group(20 mg/kg),Chan long Ding Chuan Tang Group(40 mg/kg).The normal mice were not induced by ova,the other mice were induced by ova to establish the acute asthma mice model.One week after the mice adapted to the environment,on the 0,3 and 7 days,the mice were injected with ova and emulsifier(100 μg/animal)completely emulsified with the same volume adjuvant aluminum hydroxide at multiple points on the back.On the 14 th day,ova(50 μg/animal,once a day,nose drop,7 days in total)were excited.After the model was established successfully,0.3m L normal saline was given to the model group and the normal group,and the dexamethasone group,the Chan long Ding Chuan Tang Group and the Chan long Ding Chuan Tang group were given 2 mg/kg,20 mg/kg and 40mg/kg crude drug respectively for 4 weeks.At the end of the course of treatment,the pathological changes of lung tissue were observed,and the contents of IL-6,IL-1 β,TNF-α in serum and lung tissue,as well as the contents of SOD,cat and MDA in lung tissue were measured.Western blot was used to detect the expression of Trx and Txinp oxidative stress-related proteins and the expression of MAPK and NF-κB inflammatory pathway-related proteins in lung tissues,and to investigate their possible mechanisms.Results:1.The mouse model of acute asthma was successfully replicated,and the pathological injury results showed that the modeling was successful.2.Compared with the blank control group,the content of SOD and CAT in the lung tissue of mice in the acute asthma model group was reduced(P<0.05);after drug intervention,compared with the model group,the Cianlong Dingchuan decoction group(20 mg/kg,40 mg/kg)and the dexamethasone group can increase the content of SOD and CAT in the lung tissue(P<0.05),there is no significant difference between the chanlongdingchuan decoction group and the dexamethasone group(P> 0.05).3.Compared with the blank control group,the content of MDA in the lung tissue of the acute asthma model group was significantly increased(P<0.05);after drug intervention,compared with the model group,the Cianlong Dingchuan decoction group(20 mg/kg,40 mg/kg)and dexamethasone group can reduce the MDA content(P<0.05),Chanchan Dingchuan Decoction group and dexamethasone group had no significant difference(P> 0.05).4.Compared with the blank control group,the levels of TNF-α,IL-6 and IL-1β in the serum of mice in the acute asthma model group were significantly increased(P<0.05);after drug intervention,compared with the model group,cicada dragon Dingchuantang intervention group(20 mg/kg,40 mg/kg)and dexamethasone group can reduce the content of TNF-α,IL-6,IL-1β in serum(P<0.05),Chanlong Dingchuantang There was no significant difference between the group and the dexamethasone group(P>0.05);the high-dose group(40 mg/kg)of Chanlong Dingchuan Decoction significantly reduced TNF-α,IL-6,IL-in lung tissue homogenate The content of 1β(P<0.05),the low-dose group(20 mg/kg)of Chanlong Dingchuan Decoction can reduce the content of IL-6 and IL-1β in lung tissue homogenate(P <0.05).5.Compared with the blank control group,the expressions of inflammation and oxidized proteins p-NF-κBp65,p-p38 MAPK,p-JNK,p ERK,p-IκBα,Txnip,etc.in lung tissue homogenate of acute asthma model group were significantly increased Trx protein expression was reduced(P<0.05);after drug intervention,both the chanlongdingchuan decoction group(20 mg/kg,40 mg/kg)and the dexamethasone group could reduce inflammation and oxidation-related protein p-NF-κBp65,P-p38 MAPK,p-JNK,p-ERK,p-IκBα,Txnip expression,increase the expression of antioxidant-related protein Trx(P<0.05),Cianlong Dingchuan decoction group and dexamethasone group was not significant Sex difference(P> 0.05).Conclusions:1.The preliminary results showed that CLD had a certain therapeutic effect on ovalbumin induced acute asthma mice model,and the high and low doses of CLD had a certain improvement on acute asthma lung injury.2.Chan long Ding Chuan decoction may improve acute asthma by regulating the signal pathway of txin P/TRX/MAPK/NF-κB,regulating the balance of oxidation/antioxidation and inhibiting inflammatory reaction. |
