Effects Of Chaishao Lujiao Decoction On The Behaviors And Levels Of IL-6,TNF-α And IL-10 Of Rats With Chronic Unpredictable Mild Stress-induced Depression

Objective:In this experimental research,Chaishao Lujiao Decoction was served as the main intervention measures,by observing the changes in the behavior of the depression rats with chronic unpredictable mild stress,for example,the changes of general survival,weight,the consumption of sugar water,the open-field experiments degree of change,and the serum levels of IL-6,TNF-αand IL-10.To discuss Chaishao Lujiao Decoction antidepressant effects from both macro and micro perspectives.Methods:1.Total of 48 male SD rats were selected by open-field test,the sum of the number of horizontal movement and vertical movement30-120 times.Randomly divided into seven groups: the normal group,the model group,the positive drug group,the Chaishao Lujiao Decoction high,medium,and low dose groups,with 8rats in each group.The rat model of depression was created by chronic unpredictable mild stress(CUMS)combined with solitary rearing method.2.From the first day that the model creating,the normal group and the model group were administrated with the same dose potable water,while the positive drug group were administrated with Fluoxetine hydrochloride capsule diluent and the Chinese medicine groups were administrated with different concentrations of Chaishao Lujiao Decoction once a day before accepting the stimulus.During the stress process,all of rats general survival,weight,the consumption of sugar water,the open-field experiments degree were observed.3.After 21 days of administration,and the behavioral tests were completed.Observing the IL-6,TNF-α and IL-10 level in serum of rats by enzyme-linked immunosorbent assay(ELISA).Results:1.Effect of Chaishao Lujiao Decoctionon on behaviors of CUMS depression model in rats.In the 21 st day,compared with normal group,the weight of rats in model group decreased;compared with model group,the weight of rats in Chaishao Lujiao Decoction high medium,and low dose groups increased.On the 21 st day,compared with normal group,the consumption of sugar water in model group decreased markedly(P<0.01);compared with model group,the weight of rats in Chaishao Lujiao Decoction high medium,and low dose groups increased(P<0.05).On the 21 st day,compared with normal group,the open-field experiments degree in model group decreased markedly(P<0.01);compared with model group,the open-field experiments degree in Chaishao Lujiao Decoction high medium,and low dose groups increased(P<0.05).2.Effect of Chaishao Lujiao Decoctionon on serum level of IL-6,TNF-αand IL-10 in rats with CUMS depression model.Compared with normal group,the level of serum IL-6 and TNF-α of rats in model group increased markedly(P<0.01),the level of serum IL-10 decreased markedly(P<0.01);compared with model group,thethe level of serum IL-6 and TNF-α of rats in Chaishao Lujiao Decoction high medium,and low dose groups decreased markedly(P<0.01),the level of serum IL-10 increased markedly(P<0.01).Conclusion:1.The chronic mild unpredictable stress for 21 days combined with solitary rearing method can provide an successful animal model.Chaishao Lujiao Decoction can improve the change in the behavior of the depression rats,and regulate serum level of IL-6,TNF-αand IL-10 in rats with CUMS depression model.2.Chaishao Lujiao Decoction may through regulating serum level of IL-6,TNF-αand IL-10 in rats with CUMS depression model to playing the role of antidepressant.
