Study On Mechanism Of 50% Ethanol Extracts From Erding Granule Prescription On Hperuricemia Based On Network Pharmacology

Erding granule consists of four Chinese medicines,namely Violae Herba,Isatidis Radix,Taraxacum mongolicum Hand.-Mazz and Lobelia chinensis,contained in Chinese pharmacopoeia(2015 edition)as a prescription medicine.It has the function of clearing heat and removing toxicity and is widely used in the treatment for heat boil poison,pharynx and throat swelling,wind-heat and fire eye caused by heat and toxicity.The preliminary study shows that 50% ethanol extracts of Erding granule has significant activity on lowering uric acid,but the pharmacodynamic substance is still unclear.Therefore,this study used network pharmacology to explore the pharmacodynamic substances of erding granule in the anti-hyperuricemia,and studied the content of the pharmacodynamic substances of 50% ethanol extracts from erding granule and the anti-hyperuricemia effect,mainly including the following aspects:1.The rat model of hyperuricemia was constructed by intraperitoneal injection of PO and oral administration of HX,and the anti-hyperuricemia effect of 50% ethanol extracts from Erding granule was studied.The results showed that the 50% ethanol extracts from Erding granule had a significant effect on lowering uric acid.2.Pharmacodynamic substances of Erding granule in treatment Hyperuricemia using network pharmacology.The results showed that pharmacodynamic substances,such as esculin,esculetin,caffeic acid,glycyrrhizin,luteolin,quercetin,hesperidin,kaempferoll and chicoric acid,played a role in uric acid-lowering by regulating the levels of 8 targets,including TNF-α,IL-1β,XOD,GLUT9,OAT1,ABCG2,COX-2.3.The contents of pharmacodynamic substances in the 50% ethanol extracts of erding granules was determined by ultra high performance liquid chromatography-tandem mass spectrometry.The results showed that the contents of esculin,esculetin,caffeic acid,glycyrrhizin,luteolin,quercetin,hesperidin,kaempferoll and chicoric acid were 265.84,1505.37,359.81,0.01,116.70,12.30,1.75,1.03,1643.57 μg/g extract.4.The anti-hyperuricemia effect of pharmacodynamic substances on hyperuricemia was studied based on hyperuricemia rat model.The results showed that the predicted pharmacoactive substances had significant uric acid-lowering effects,and could inhibit the levels of XOD,TNF-α,IL-1β,as well as the expression of GLUT9 and increase the expression of OAT1.Thus,the pharmacodynamic substances in the 50% ethanol extracts from Erding granule mainly include esculin,esculetin,caffeic acid,glycyrrhizin,luteolin,quercetin,hesperidin,kaempferoll and chicoric acid.The main pharmacodynamic substances can reduce the levels of XOD,TNF-α,IL-1β,as well as the expression of GLUT9 and increase the expression of OAT1.
