Clinical medicine of traditional Chinese medical takes the form of compound through the compatibility between different drugs use.Because the characteristic of traditional Chinese medicine is that they have contained complicated chemical composition and more interference factors,quality control has been the difficulties of the study of Traditional Chinese medicine preparation.Qiteng Xiaozhuo Granule is the prescription which based on the clinical experience of the famous expert Cao Enze who is working in the Renal medicine of the first affiliated hospital of Anhui university of traditional Chinese medicine.The prescription is consist of Triptergium wilfordii,Astragali Radix,Hedyotis diffusa Willd,Coix lacryma-jobi L.var.mayuen(Roman.)Stapf,Imperata cylindrica Beauv.var.major(Nees)C.E.Hubb,Cryptotympana pustulata Fabricius,Leonurus japonicus Houtt.Up to now,never happen adverse reactions.In the early research,Astragalus and Hedyotis diffusa was indentified by thin-layer chromatography,the content of Stachydrine was determinated and establishing a method of Wilforlide A limit test by TLC method.This experiment is on the basis of previous research.We identified five drugs by thin-layer chromatography.The content of three active ingredients were determinated by High-performance liquid chromatography.The quality of preparation was controlled effectively by these methods.Even though Triptergium wilfordii Hook.f.and Traditional Chinese medicine preparation which has Triptergium wilfordii Hook.f.has very remarkable efficacy as the treatment of chronic glomerulonephritis they also have obvious side effects.Three referential indexes were selected which including Triptolide,Celastrol and Wilforlide.We study the change of the content of these main referential indexes by compare Tripterygium Wilfordii with Qiteng Xiaozhuo Granule.We take the study of the detoxicity effect of Tripterygium Wilfordii by compatibility mechanism on chemical components.All these are to improve the security and to limit the toxicity of granule which has Tripterygium Wilfordii in it.The granule was applied more widespread based on the research.Objective: 1.To improve the quality standard of Qiteng Xiaozhuo Granule.2.To establish the measure method of triptolide,celastrol and wilforlide in Tripterygium wilfordii and Qiteng Xiaozhuo Granule.We want to explore the detoxicity effect of Tripterygium wilfordii by compatibolity mechanism on the base of comparing the change of content.Methods: 1.On the basis of perliminary study,we consult literature materials and screened the best identify method of Coicis Semen,Imperatae Rhizome,Cicadae Periostracum and Leonurus Japonicus by thin-layer chromatography.2.Study chromatogram conditions,establish the measure method of astragaloside ? by using UPLC-ELSD method and establish the measure method of calycosin and oleanolic acid by using UPLC method.3.To establish the measure method of triptolide,celastrol and wilforlide by using UPLC method.Then detect the content of these referential indexes in Tripterygium wilford and Qiteng Xiaozhuo Granule.Compare the changing of the amount of triptolide,celastrol and wilforlide to explore the basis chemical components about detoxicity effect of tripterygium wilfordii by compatibolity mechanism in Qiteng Xiaozhuo Granule.Results: 1.According to literature material and constantly experiment the TLC of Coicis Semen,Imperatae Rhizome,Leonurus Japonicus,Cicadae Periostracum and Teipterygium Wilforlide show that these spots were clear without interference.2.On the basis of experiment all separation steps were carried out on an Agilent Eclipse plus C18 column(4.6mm×250mm,5?m)we establish an accurate and reproducible content method which can measure the content of astraga losiwhde?,calycosin and oleanolic acid.3.We establish a very effective method for the content of Triptolide,Celastrol and Wilforlide.The statistics analysis showed that the content of triptolide,celastrol and wilforlide in Qiteng Xiaozhuo Granule reduced significantly which compared with tripterygium wilford.The result show that Tripterygium wilfordii has detoxification after the formula.Conclusion: The quality of Qiteng Xiaozhuo Granule can be improved effectively by the methods of thin-layer chromatography and ultra high performance liquid chromatography.The method is reliable and repeatability.The toxicity of Tripterygium wilfordii was reduced significantly by compatibolity mechanism by analyze the date.
