| ObjectiveIn this study,cisplatin(DDP)model rats were used as the research object,acupuncture combined with acupoint application as the treatment method,Zhongwan,Neiguan and Zusanli as the treatment prescription,through observing the basic quality of life of rats,recording the intake of kaolin(i.e.foreign body)to monitor the vomiting response and the degree of vomiting response.Then the expression of 5-HT3 R gene was detected by PCR and the contents of ACh and NOS were detected by immunohistochemistry,in order to explore the mechanism of acupuncture combined with Acupoint Application on improving the adverse reactions of gastrointestinal tract after chemotherapy.Methods60 Sprague Dawley male rats were randomly divided into blank group,model group,acupoint application group(Acupoint Application Group),acupuncture group and acupuncture plus Acupoint Application Group(acupuncture + acupoint group).Wu Zhuyu Decoction and Wen Dan Decoction were used to extract the effective ingredients of the medicine and make the plaster.The blank group was given 5 mg / kg intraperitoneal injection of normal saline,the other four groups were given 5 mg / kg intraperitoneal injection of DDP to create the model of gastrointestinal adverse reactions after chemotherapy,and the model was completed 24 hours later.Point sticking group,acupuncture group and acupuncture + point group were treated with "Zhongwan","Neiguan" and "Zusanli" respectively.Once a day for 7 days.The rats were killed after fasting for 1 day,and the whole stomach and ileum were taken out for detection.The expression of 5-HT3 R gene and the content of ACh and NOS in gastrointestinal tissues of rats in each group were detected,and the differences between each group were compared.Results1.The experiment of rat heterophagia.The intake of kaolin in rats reflected the vomiting of rats: after intraperitoneal injection of cisplatin,compared with the blank group,the intake of kaolin in each group increased significantly in 12 hours and 24 hours after the establishment of the model(P < 0.01);after 7 days of treatment,compared with the model group,the total intake of kaolin in acupoint sticking group,acupuncture group and acupuncture + acupoint group was less than that in the model group The difference was statistically significant(P < 0.01);the most significant difference was in the acupuncture + point group,but there was no statistical significance compared with the point sticking group and the acupuncture group(P > 0.05).2.Compared with the blank group,the expression of 5-HT3 R in the gastric tissue of the model group was significantly higher(P < 0.01);after 7 days of treatment,compared with the model group,the expression of 5-HT3 R in the gastric tissue of the acupoint sticking group,the acupuncture group and the acupuncture + acupoint group was significantly lower(P < 0.01).Compared with acupoint sticking group and acupuncture group,the expression of 5-HT3 R in stomach tissue of acupuncture + acupoint group was lower,but there was no statistical significance(P > 0.05).3.Compared with the blank group,the expression of ache in the gastric tissue of the model group was significantly higher,and the difference was statistically significant(P < 0.01).After 7days of treatment,compared with the model group,the expression of ache in the gastric tissue of acupoint sticking group,acupuncture group and acupuncture + point group was significantly lower,the difference was statistically significant(P < 0.05,P < 0.01),among which the acupuncture + point group was of great significance(P < 0.01).Compared with acupoint sticking group and acupuncture group,the expression of ache in gastric tissue of acupuncture + acupoint group decreased,but there was no statistical significance(P > 0.05).4.Compared with the blank group,the expression of NOS in intestinal tissue of model group was significantly lower(P < 0.01).After 7 days of treatment,compared with the model group,the expression of NOS in intestinal tissue in the acupoint sticking group,acupuncture group and acupuncture + acupoint group increased significantly,the difference was statistically significant(P < 0.01).Compared with acupoint sticking group and acupuncture group,the expression of NOS in intestinal tissue of acupuncture + acupoint group increased,but there was no statistical significance(P > 0.05).Conclusion1.Acupuncture combined with acupoint application can effectively improve their vomiting symptoms and gastrointestinal adverse reactions,and improve their quality of life.2.Acupuncture and acupoint application can effectively improve the adverse reactions of gastrointestinal tract after chemotherapy.Their effects and mechanisms are similar.The two methods can be used alternately,providing more means for clinical treatment.3.The effect of acupuncture combined with acupoint application is better than that of acupuncture and acupoint application alone,which has the best effect and can be widely used in clinic.4.Acupuncture combined with acupoint application makes rat the excitatory neurotransmitter in gastrointestinal tissue The expression of 5-HT3 R and ache decreased,the expression of inhibitory neurotransmitter NOS increased,so that the neurotransmitters which were checked and balanced could be regulated,so as to effectively improve the adverse reactions of gastrointestinal tract after chemotherapy.The regulation of neurotransmitters by acupuncture combined with acupoint sticking may be one of the mechanisms to improve the adverse reactions of gastrointestinal tract after chemotherapy. |
