| Oral squamous cell carcinoma(OSCC)is one of the most common cancers in oral and maxillofacial region.Solute carrier family 3 member 2(SLC3A2)regulates calcium levels and transports 1-type amino acids,which plays a certain role in the development of tumors.Research objectives:1.To observe the expression of SLC3 A2 in oral squamous cell lines and study its effect on the function of oral squamous cell cells.2.The expression of SLC3A2 in oral squamous cell carcinoma was observed to study its effect on the prognosis of patients with oral squamous cell carcinoma.Research methods:1.The expression of SLC3A2mRNA in 3 oral squamous cell cells and 1 normal epithelial tissue cell was detected by QRT-PCR(real-time PRC).2.Plasmid of SLC3A2 was constructed,and the expression of SLC3A2 was down-regulated by transfection of lipofectamine3000 into oral squamous cell cells.3.The effect of down-regulation of SLC3A2 expression on the proliferation of HN6 in oral squamous cell carcinoma cells was detected by CCK8 method.4.Transwell invasion and migration assay was used to detect the effect of down-regulation of SLC3A2 expression on the invasion and migration of HN6 in oral squamous cell carcinoma cells.5.The effect of down-regulation of SLC3A2 expression on the apoptosis of HN6 in oral squamous cell carcinoma cells was detected by flow cytometry.6.The expression of SLC3A2 in 96 oral squamous cell carcinoma tissues and adjacent tissues was detected by immunohistochemistry.Research results:1.Compared with the normal epithelial cell group,SLC3A2 of oral squamous cell carcinoma group was highly expressed(P<0.01).2.The results of CCK8 method showed that the proliferation ability of oral squamous cell carcinoma was inhibited after the expression of SLC3A2 was down-regulated(P<0.05).3.Experimental results of Transwell invasion and migration showed that the invasion and migration ability of oral squamous cell carcinoma were inhibited after SLC3A2 expression was down-regulated.4.Flow cytometry showed that the apoptosis rate of oral squamous cell carcinoma increased after the expression of SLC3 A2 was down-regulated.5.Immunohistochemical results showed that SLC3A2 was mainly expressed on the cell membrane of oral squamous cell cells,and the expression in oral squamous cell carcinoma tissues was higher than that in normal epithelial tissues(P<0.05).6.The positive expression of SLC3A2 in oral squamous cell carcinoma was not related to age,sex,or lymph node metastasis(P>0.05),but was related to the grade of the tumor(P<0.05).Conclusion:SLC3A2 is highly expressed in the tissues of oral squamous cell carcinoma,mainly expressed on the cell membrane of oral squamous cell carcinoma,which may promote the proliferation,invasion and migration of oral squamous cell carcinoma and inhibit the apoptosis of squamous cell carcinoma,and is expected to become a tumor marker of oral squamous cell carcinoma. |
