Objective: By detecting the expression of methyltransferase-like protein 16(METTL16)in gastric cancer,exploring the relationship between the expression level of METTL16 and the clinicopathological characteristics of patients with gastric cancer,aiming to explore the expression level of METTL16 in gastric cancer Clinical significance.Methods: The difference data of METTL16 in different cancer and para-cancerous tumor samples was extracted from the TCGA database,and then combined with the METTL16 expression data obtained in GTEx database in 31 different source tissues,and finally the expression differences of METTL16 in 27 tumors were obtained.Immunohistochemical technique was used to detect the expression level of METTL16 in gastric cancer and its adjacent tissues,and analyze its relationship with the clinical data of patients.The GEPIA database analyzes the overall survival rates of gastric cancer patients at different levels of METTL16 expression,followed up patients with positive METTL16 expression,and analyzed the factors affecting the survival and prognosis of patients.Results:(1)The expression level of METTL16 was higher in cholangiocarcinoma,esophageal carcinoma,and polymorphonuclear gliomas,and higher in bladder urothelial carcinoma,cervical squamous carcinoma,adenocarcinoma,and paracancerous tissues such as colon carcinoma(P<0.05).(2)The positive expression level of METTL16 in patients with gastric cancer was significantly higher than that in adjacent tissues(P<0.001).(3)The positive expression level of METTL16 is related to the degree of differentiation(P=0.002),the degree of invasion(P<0.001),tumor stage(P<0.001)and the presence or absence of lymph node metastasis(P<0.001)in patients with gastric cancer.(4)The degree of differentiation(P=0.001),degree of invasion(P=0.020),tumor stage(P=0.001)and the expression level of METTL16(P=0.023)of patients with gastric cancer are related to the patients The survival and prognosis are related,and the degree of tumor differentiation(OR=0.262,95%CI=0.078~0.875)is an independent factor affecting the survival and prognosis of patients(P=0.029).(5)In the clinical stage III~IV,the 3-year survival rate of gastric cancer patients with high expression of METTL16 is lower than that of patients with low expression(P=0.025).Conclusions:(1)METTL16 is significantly high expression in gastric cancer tissues.(2)The higher the positive level of METTL16,the lower the degree of differentiation of gastric cancer,the stronger the invasiveness,the later the tumor stage,and the easier it is to metastasize to lymph nodes.(3)The higher the positive level of METTL16,the lower the degree of differentiation,the stronger the invasiveness,and the later the clinical stage of gastric cancer patients have a poorer survival prognosis.Among them,the degree of tumor differentiation is an independent factor affecting the survival and prognosis of patients.(4)For patients with gastric cancer in the tumor stage III-IV group,the higher the level of METTL16 positive,the worse the prognosis of gastric cancer.
