| Objective This study intends to use the traditional Chinese medicine inheritance support system(3.0)to systematically sort out and excavate Professor Song Min medication experience and the characteristics of treatment rules and treatment methods for the treatment of lumbar disc herniation,and to obtain the experiential prescriptions through calculation-assisted extraction.Then based on network pharmacology,molecular docking and other means to predict and discuss the molecular mechanism of the empirical formula in the treatment of lumbar intervertebral disc herniation,establish an integrated visual network,study its mechanism of action from a multi-faceted and multi-dimensional relationship,and then theoretically Verification of the efficacy and mechanism of the predicted "experience prescription" will provide reference and basis for the future development of cell or zoology experimental verification and the development of new drugs for the treatment of lumbar disc herniation with traditional Chinese medicine.Methods A review of Professor Song Min outpatient prescriptions for the treatment of lumbar intervertebral disc herniation from September 2018 to December 2020.With reference to the inclusion and exclusion criteria,309 prescriptions were sequentially entered into the TCM inheritance support system(3.0)system,and the statistics provided by the platform were used,Correlation,cluster analysis and other functions,summarize the characteristics of Professor Song Min treatment of lumbar intervertebral disc herniation,integrate and summarize the characteristics of his medication,classify the core drug pairs and corner drugs,and refine and summarize the experience.The second step is to use open platforms such as TCMSP,BATMAN,Swiss target prediction,and combine with Pub Med literature reports to obtain the active ingredients of the "experience formula" and the corresponding target protein;Cytoscape 3.4.0 software builds a visual "drug-active ingredient-target" Network diagram;Retrieve lumbar disc herniation related targets through Gene Cards and OMIM;map drug targets and disease targets(Veen Diagram.R)to obtain potential targets of experience prescriptions for treatment of lumbar disc herniation;map the obtained targets Transfer to STRING 11.0 database for PPI analysis;use DAVID and omicshare network platforms for GO and KEGG enrichment analysis of the mapped common targets;finally use Zinc database,Auto Dock,and Py MOL software for molecular docking of main compounds and targets Validation,so as to find out the key targets and signal pathways of drug treatment of diseases,so as to explore the mechanism of the empirical prescription for the treatment of lumbar disc herniation.Result Through the analysis of 309 outpatient prescriptions,a total of 156 types of medicines were involved.The top 15 Chinese medicines used in frequency were Angelica(290),Chuanxiong(259),Astragalus(252),Eupatorium(251),and Licorice(220).),Sanleng(189),Curcuma(188),Red Peony(171),Earth Dragon(136),Turmeric(122),Papaya(119),Eucommia(114),Shichangpu(114),Weilingxian(113),Achyranthes bidentata(113);the four-qi of the drug ranked the most frequently in the temperature medicine(2282),followed by the flat medicine(848),the cold medicine and the cold medicine were used less frequently;in the five taste analysis,the bitter medicine was the most common(2219),followed by spicy medicines(2073),sweet medicines with frequency(1688),bitter,pungent,and sweet medicines have little difference in frequency,salty medicines and sour medicines are used less frequently;the distribution shows that most medicines The two meridians of the liver and the spleen,the heart,lung,stomach,and kidney are involved;the results of efficacy statistics show that the drugs for promoting blood circulation and removing blood stasis(1395),tonic(1075)and dispelling rheumatism(466)are commonly used drugs.According to the drug association rules,five core drugs were counted,namely Angelica,Licorice,Chuanxiong,Astragalus,and Eupatorium.Angelica –Glycyrrhiz a-Astragalus-,Astragalus-Eupatorium-Angelica,Chuanxiong-Astragalus-Angelicaarecommonly used drug combinations.The three cluster analysis results showed that the combination frequency of Angelica,Astragalus,Eupatorium,Ligusticum chuanxiong,Licorice,and Eucommia was the highest and the classification overlap was the least,representing the clearest classification and the most obvious clustering.145 active ingredients of the "experience formula" and 323 corresponding targets were obtained by searching,and the Cytoscape 3.4.0 software draws the visual display of the disease targets corresponding to the obtained "experience formula" active ingredients,constructing a "drug-component-target" "Dot" network diagram,showing that quercetin,kaempferol,and 7-O-methylisoxylitol are important active ingredients of the "experience formula";381 LDH-related genes were obtained;52 common targets were obtained after mapping,PPI It shows that IL-6,ALB,TNF,AKT1,MMP9,VEGFA,PTGS2,MAPK1,etc.may be the core target of the empirical prescription to treat LDH.GO enrichment analysis includes biological processes including reactions to external stimuli,reactions to oxygenated compounds,lipopolysaccharides,and bacteria-derived molecules;signal receptor binding,cytokine receptor binding,enzyme binding,molecular function modifiers and other molecules Function;cell components such as membrane microzone,extracellular space,and extracellular area.KEGG pathway enrichment analysis identified more than 20 signal pathways including TNF,MAPK,PI3K-Akt,NF-κB,Toll-like,HIF-1,osteoclast differentiation,and estrogen.Molecular docking shows that the main active components of the "experience formula",quercetin,kaempferol,etc.,have good binding properties with core targets such as TNF,IL-6,PTGS2,and MAPK1(derived from the PPI network topology).In conclusion 1.Professor Song Min treatment of lumbar intervertebral disc herniation is based on strengthening the body and eliminating the evils as the main treatment principles.The treatment method is mainly to replenish qi and activate blood,replenish the liver and kidney,and relax the tendons and collaterals.At the same time,he pays attention to combing the Qi machine and regulating the spleen and stomach.Traditional Chinese medicines for promoting blood circulation,removing blood stasis,tonic and dispelling rheumatism are often used in treatment.According to association rules and cluster analysis,it is found that Professor Song Min experience in treating lumbar disc herniation is Angelica,Astragalus,Eupatorium,Chuanxiong,Licorice,The drug combination of Eucommia.2.The empirical prescription mainly uses 133 active ingredients such as quercetin,kaempferol,7-O-methylisoxylitol to act on 52 key targets such as IL-6,TNF,MMP9,VEGFA,PTGS2,MAPK1,etc.,and regulate Multiple biological functions and signal pathways,so as to play a role in the treatment of lumbar disc herniation. |
