Objective: By measuring the plasma PT,APTT,TT,FIB,DD levels of rats in the experimental group and the control group,to explore the coagulation function of rats under hypoxia;by comparing the mRNA levels of coagulation factors in the liver tissues of the experimental group and the control group,to explore the level of coagulation factors in the liver tissue of rats under hypoxia,and to provide some theoretical basis for abnormal blood coagulation under hypoxic environment.Methods: Twenty male SD rats were selected as the research objects,and randomly divided into 2 groups.The experimental group was raised in a hypobaric oxygen chamber at a simulated altitude of 5000 meters,and the control group was raised in Xining(2300 meters).The two groups of rats eat the same amount of water and food each day.Consistent,30 days later,intraperitoneal injection were used to anesthetiz the rats,and the arterial blood and liver tissues of the rats were collected,and the WBC,RBC,Hb,HCT,PLT levels of the rats were measured by the automatic five-category animal blood cell analyzer,the levels of PT,APTT,TT,FIB in rats were determined by the automatic blood coagulation analyzer,and the levels of DD in rats were analyzed by an enzyme label analyzer.The mRNA expression levels of FII,FVII,FVIII,FIX,FX,FXI,FXII,FXIIIA subunits in rat liver tissues were determined by fluorescence quantitative PCR method.Results:Compared with the control group,the experimental group increased the levels of RBC,Hb,and HCT(P<0.001),and the count of PLT was decreased(P<0.001).the levels of PT,FIB,and DD was increassed in the experimental group(P<0.05).There was no difference in the levels of TT and APTT between the two groups(P>0.05).The relative expression of FII,FVII and FXI mRNA in the liver tissues of the experimental group was higher than that of the control group(P<0.05);while the relative expression level of FVIII mRNA in liver tissues of the experimental group was lower than that of the control group(P <0.05);the relative expression levels of FIX,FX,FXII,and FXIIIA subunits in liver tissues of the experimental group had no difference compared with the control group(P>0.05).Conclusion:(1)Rats’ RBC,Hb,and HCT levels increase under hypoxia,which may be a compensatory response of rats to hypoxia;(2)Rats’ PLT levels under hypoxia decrease,and FIB and DD levels increase,prolonged PT,considering that hypoxia may activate the coagulation system of rats and increase the risk of thrombosis under hypoxia;(3)The synthesis of FII,FVII,FXI mRNA in rat liver under hypoxia increases;and the mRNA of FVIII in rat liver tissue under hypoxia synthesis is reduced.
