| Objective:To observe the effect of Jianpi Huazhuo Jiangzhi Decoction on obesity mice model,adipokines,pro-inflammatory cytokines,blood lipids and blood glucose and other related indicators,pathomorphology and Changes of PPARγ/SREBP-1c/TNF-α/IL-6 mRNA and protein expression,to investigate the effect of this prescription on metabolic homeostasis and chronic inflammation in obese mice and its mechanism.Methods:Establishment of obesity mouse model induced by high fat diet,the traditional Chinese medicine group was given Jianpi Huazhuo Jiangzhi Decoction and the western medicine group was given orlistat capsule,implementation of group control treatment.1.During the treatment period,the body weight,abdominal circumference,mental state and activity of mice were measured and recorded;2.At the end of treatment,the distribution,content and volume of fat were detected by micro CT imaging in vivo;3.Observe the shape of liver,weigh the wet weight of liver,visceral fat and subcutaneous fat,and calculate the corresponding coefficient;4.Serum lipid profile related indicators,blood glucose indicators,automatic biochemical analyzer was used;5.Serum leptin,resistin,adiponectin,insulin,interleukin-6,tumor necrosis factor-α and free fatty acid were detected by ELISA;6.The pathomorphological structure of liver and adipose tissue was observed by light microscope;7.TNF-α / IL-6 mRNA expression was detected by RT q PCR;8.The protein expression of PPAR γ / SREBP-1c was detected by Western blot;9.Statistical software SPSS was used to record and analyze the experimental data.Results:1.General observation and weight change: the mental state,activity,skin gloss and the degree of reaction to the outside world of the mice in the experimental groups were improved,which were better than those in the model group.Among them,high dose of Jianpi Huazhuo Jiangzhi Decoction has the best effect,the weight of mice in high dose group decreased significantly,good mental state,increased activity,glossy fur,sensitive response to the outside world,normal feces,followed by the middle dose group;2.Fat volume,content and distribution: compared with the model group,the visceral fat and total fat volume of the experimental groups were significantly reduced,while the subcutaneous fat was reduced at the same time by high dose,with statistical difference(P <0.05).The results showed that all the treatment groups could effectively reduce the content and volume of visceral fat,and the curative effect was similar;the comparison of the content and range of fat distribution showed that the treatment groups mainly acted on visceral fat to reduce the distribution of visceral fat,and the high-dose treatment had the best curative effect,followed by the middle dose and Western medicine,and the low-dose treatment was relatively not obvious;3.The effect on blood lipid and blood glucose: the high dose group and Western medicine group in reducing TC,TG,LDL-C levels,increasing HDL-C levels,the results were statistically significant(P < 0.05),and the curative effect was the same;in reducing Glu and apo B,the high dose group had the best curative effect,followed by western medicine group.The therapeutic effect of Jianpi Huazhuo Jiangzhi Decoction in three gradient dose groups was in the order of high dose,medium dose and low dose;4.Effect on adipokines: Jianpi Huazhuo Jiangzhi Decoction has the same effect as western medicine in reducing the concentration of leptin,resistin,insulin and increasing the concentration of adiponectin.Among the three gradient doses of traditional Chinese medicine,high dose has the best effect,followed by medium dose,and low dose has the weakest effect;5.Effect on pro-inflammatory cytokines: the concentrations of IL-6,TNF-α and FFA in serum of all groups were effectively reduced,and the results were statistically significant(P <0.05);6.Effect on liver morphology and wet weight: the liver morphology,texture and color could be restored to different degrees in all groups,and the high dose was the best,followed by the medium dose,and the western medicine and low dose were the best.The wet weight of liver and the volume of liver were reduced in all groups,and the results were statistically significant(P < 0.05);7.Effect on wet weight and coefficient of visceral fat and subcutaneous fat: The wet weight and coefficient of visceral fat and subcutaneous fat in each treatment group were decreased in varying degrees,with statistical difference(P < 0.05).The results showed that high dose of the Chinese medicine group had the most obvious effect and the best effect in reducing the wet weight and content of visceral fat;8.Pathological observation of liver and adipose tissue: Jianpi Huazhuo Jiangzhi Decoction high dose group had the best recovery of liver morphology and structure,significantly reduced the number of yellow lipid droplets vacuoles in cytoplasm,recovered the middle position of nucleus,and the size of hepatocytes was uniform,followed by western medicine and medium dose;the volume of adipocytes in each group was reduced in varying degrees,and the number of adipocytes in unit field of vision was increased,among which the high-dose of Chinese medicine was the most effective,while the low-dose group could be seen Inflammatory cell infiltration;9.Effect on the expression of TNF-α and IL-6 mRNA: the relative expression of TNF-αand IL-6 mRNA in adipose tissue could be effectively regulated in all groups,with statistical difference(P < 0.05);10.Effect on PPAR γ and SREBP-1c protein expression: the experimental groups can effectively down regulate the protein expression of SREBP-1c gene in liver and adipose tissue and up regulate the protein expression of PPAR γ gene,with statistical difference(P < 0.05).The high-dose Chinese medicine group has the best curative effect,followed by the medium dose and Western medicine.Conclusion:1.Jianpi Huazhuo Jiangzhi Decoction has significant anti obesity effect,and can significantly reduce blood lipid and blood glucose,regulate adipokines and pro-inflammatory cytokines,maintain serum adiponectin level,reduce local inflammation level and improve metabolic disorder,so that the body is in a healthy metabolic steady state.2.Jianpi Huazhuo Jiangzhi Decoction can effectively reduce the liver injury induced by HFD and restore the morphological structure of liver and adipose tissue.3.The mechanism of Jianpi Huazhuo Jiangzhi Decoction improving metabolic disorder and chronic inflammation in obese mice may be up regulating PPAR γ gene and down regulating SREBP-1c gene protein expression,down regulating TNF-α and IL-6 gene mRNA relative expression,so as to produce the effect of regulating metabolic homeostasis and inflammation.In conclusion,Jianpi Huazhuo Jiangzhi Decoction has the best effect in the three gradient doses,followed by the middle dose and the low dose.The recovery of spleen and stomach function,it can effectively regulate physiological fat production and metabolic health,and provide basis and theoretical support for clinical practice. |
