Objective: The expression of B-Raf,MEK and ERK in reflux esophagitis and Barrett esophagus were detected to explore the role of ERK signal channels in reflux esophagitis.Methods: 40 cases of reflux esophagitis tissues,39 cases of Barrett esophagus tissues and 8 cases of normal esophageal mucosal tissues were collected.The expression levels of B-Raf,MEK,ERK proteins in esophageal tissues of the three groups were detected by immunohistochemistry,and q RT-PCR technique was used to detect the expreesion levels of B-Raf,MEK,ERK proteins in 17 reflux esophagitis tissues and its adjacent normal esophageal tissues.To compare the correlation among B-Raf,MEK and ERK.Results: 1.There were 79 patients with reflux esophagitis and Barrett esophagus,the average age were 54.96±15.08 years old,30 patients were female(38.0%),49 were male(62.0%),there were 13cases(16.5%)of H.pylori infection.The symptoms of abdominal distension or abdominal pain were 31(39.2%)cases,primarily heartburn or reflux were 27(34%)cases,abdominal distension or abdominal pain in 22(52.3%)cases,dysphagia or obstruction were 10(12.7%)cases,chest pain were 3(3.8%)cases and others were 8(10.1%)cases.2.The results showed that B-Raf MEK and ERK were expressed in normal esophageal mucosal tissues,reflux esophagitis and Barrett’s esophagus,but their expressions were extremely low in normal esophageal mucosal tissues,mainly expressed in the cytoplasm.In normal esophageal mucosal tissues,reflux esophagitis tissues and Barrett’s esophagus tissues,their expression level was gradually increased(P < 0.05).Real-time fluorescence quantitative PCR showed that m RNA expression were up-regulated in the tissues of reflux esophagitis,and there were significant differences compared with the adjacent normal esophageal tissues(P < 0.05).3.The expression of B-Raf in patients with reflux esophagitis and Barrett’s esophagus was not correlated with age,gender,smoking,drinking,H.pylori(HP)infection and grade of reflux esophagitis(P > 0.05).MEK was not correlated with gender,age,H.pylori infection and grade of reflux esophagitis(P > 0.05),but was correlated with smoking and dringking(P < 0.05).The expression of ERK was not associated with gender,sex,age and H.pylori infection(P > 0.05),but was associated with smoking,drinking and grade of reflux esophagitis(P < 0.05).Conclusion: 1.Reflux esophagitis and Barrett esophagus occur in middle-aged men,which is mainly manifested by abdominal distension or abdominal pain and heartburn or reflux and may be related to smoking and drinking.2.The expression of B-Raf,MEK and ERK is up-regulated in normal esophageal mucosal tissues,reflux esophagitis tissues and Barrett esophagus tissues.These results indicate that ERK signaling pathway may be involved in the occurrence of reflux esophagitis,especially in the occurrence of Barrett’s esophagus.3.The expression levels of m RNA of B-Raf,MEK and ERK protein in reflux esophagitis tissues are higher than that in their adjacent normal esophageal tissues.Their abnormal expression may be involved in the development of reflux esophagitis.
