| Objective: Immune-related lncRNA related to the prognosis of cervical cancer were determined by TCGA database to construct a prognosis model of cervical cancer.Methods:Cervical cancer trancriptome,clincal data and immune-related gene sets were downloaded from the TCGA Database and Molecular Signature Database.The immunogene-lncRNA co-expression was analyzed according to the immunogene-related gene set and cervical cancer transcriptional data to screen the immunogene-related lncRNA(cor>0.4,P<0.001).Univariate COX regression screened out lncRNAs related to the prognosis of cervical cancer.Multivariate COX regression analysis was used to construct lncRNA model,and AIC values were used to determine the best model.The risk value of each patient was calculated according to the model,and patients were divided into high and low risk groups according to the median.K-M survival analysis was performed for the two groups and the model was evaluated by ROC.K-M survival analysis was performed between the confirmed lncRNAs.The lncRNAs with P<0.05 in the multivariate COX and K-M survival analysis were selected for function prediction.The encoding protein genes associated with lncRNA was identified by co-expression analysis.GO enrichment analysis and KEGG pathway enrichment analysis were used to predict the function of lncRNA.Results:(1)637 immune-related lncRNAs were screened out by constructing of immune-lncRNA co-expression network;(2)34 immune lncRNAs related to cervical cancer prognosis were further identified by Univariate COX regression analysis;(3)Multivariate COX regression and AIC values were used to determine 8 lncRNAs to construct the most accurate model.The risk value of each patient was calculated according to the model,and the patients were divided into high risk group and low risk group according to the median value.K-M survival analysis showed that the patients in the high risk group had poor prognosis.The ROC curve verified the model,and the area under the curve was 0.758;(4)In multivariate COX and K-M survival analysis,the lncRNAs with P<0.05 were AC004847.1,AC099568.2 and ATP21-AS1;(5)GO enrichment analysis suggested that the functions of AC004847.1 and ATP21-AS1 might be related to immune receptor activity and NADH dehydrogenase activity,respectively;KEGG enrichment analysis indicated that AC004847.1 was mainly involved in T cell receptor signaling pathway.ATP21-AS1 is mainly involved in the oxidative phosphorylation pathway.The GO and KEGG enrichment analysis of AC099568.2 showed no results.Conclusion:The prognostic evaluation model for cervical cancer was constructed based on 8 key immune-related lncRNAs,which had better effect than other clinical indicators.The main functions of AC004847.1and ATP21-AS1 were immune receptor activity and NADH dehydrogenase activity,respectively.The major pathways involved were the T cell receptor signaling pathway and the oxidative phosphorylation pathway. |
