Objective:To observe and discuss the improvement of motor function and brain tissue morphology of cerebral ischemia rats by Acupuncture-rehabilitation Treatment and the regulation of gap junction communication in the brain tissue of ischemic penumbra area.Providing experimental theoretical support for the clinical application of the acupuncture-rehabilitation method and the therapeutic target of cerebral ischemia.Method:120 male SPF Sprague-Dawley(SD)rats were randomly divided into 5 groups,namely,sham operation group,model group,acupuncture group,rehabilitation group,and acupuncture-rehabilitation group.Each group was divided into equal parts for 3d and 7d.There are 2 subgroups(n=12).The last four groups blocked the middle cerebral artery blood flow in rats by a modified intravascular suture method to simulate permanent cerebral ischemia.The modeling process of the sham operation group did not include the insertion of sutures.The sham operation group and the model group were not treated,the acupuncture treatment group was treated with scalp acupuncture,the rehabilitation treatment group was treated with treadmill exercise training,and the acupuncture treatment group was treated with two treatment methods at the same time.Rats were scored using mNSS and Zea-longa at two preset time points after treatment;the heads were decapitated immediately after scoring,and HE staining was used to observe the morphological changes of the brain tissue in the ischemic area under the electron microscope;Western Blot was used to detect and analyze the expression changes of Cx43,P-Cx43 and PKC in the ischemic penumbra of rat brain tissue.Result:1.mNSS score and Zea-longa score: 3d and 7d after operation,the scores of each group were higher than those of the sham operation group(P<0.05);3d after the operation,the scores of the acupuncture-rehabilitation group were lower than the model group(P<0.05);Compared with the model group,the scores of the acupuncture group and the rehabilitation group were reduced,but there was no significant difference(P>0.05);7 days after the operation,the scores of each treatment group were reduced compared with the model group(P<0.05)and the score of the acupuncture-rehabilitation treatment group was reduced compared with the acupuncture treatment group and the rehabilitation treatment group(P<0.05).2.HE staining: 3d and 7d after the operation,the nucleus and cytoplasmic structure of the brain tissue in the acupuncture group and rehabilitation group were clearer,the number of nerve cells increased compared with the model group,the degree of nuclear pyknosis decreased,and the cytoplasm was slightly edema,The gap becomes smaller,and the number of inflammatory cells and glial cells is also significantly reduced.On the 3rd and 7th day after the operation,the nerve cells of the acupuncture-rehabilitation group were reduced compared with the sham operation group,but compared with the model group,acupuncture group and rehabilitation group,the nucleus structure was relatively complete,the cytoplasm and microvascular edema were reduced,and the intercellular space was basically back to normal.3.The expression of Cx43 protein in brain tissue: 3d and 7d after operation,the expression of Cx43 protein in rat brain tissue was up-regulated in the model group compared with the sham operation group(P<0.05);3d after operation,the acupuncture-rehabilitation treatment group and the rehabilitation group In rats,the expression of Cx43 protein in the brain tissue of rats was significantly lower than that in the model group(P<0.05).The expression of Cx43 protein in the acupuncture group was also lower than that in the model group,but there was no statistical difference;7 days after the operation,each treatment group was compared with the model group,the expression of Cx43 protein decreased significantly(P<0.05);and at the two time points,compared with the acupuncture group and the rehabilitation group,the expression of Cx43 protein in the brain tissue of the acupuncture-rehabilitation group was significantly decreased(P<0.05).4.The expression of P-Cx43 and PKC protein in brain tissue: 3d and 7d after the operation,compared with the sham operation group,the expression of P-Cx43 and PKC protein in the brain tissue of the model group decreased significantly(P<0.05);After 3 days,the expression level of P-Cx43 protein in the brain tissue of rats in each treatment group was higher than that in the model group(P<0.05),and the expression level of PKC protein in the brain tissue of rats in each treatment group was higher than that in the model group,but not significantly difference(P>0.05);After 7days,the expression levels of P-Cx43 and PKC protein in the brain tissue of rats in each treatment group were higher than those in the model group(P<0.05);at 2 time points after treatment,the acupuncture-rehabilitation group was similar to the acupuncture group and the rehabilitation group.In contrast,the expression levels of P-Cx43 and PKC protein in rat brain tissue were significantly increased(P<0.05).Conclusion:1.Acupuncture-rehabilitation Treatment can reduce the mNSS score and Zea-longa score of rats with cerebral ischemia,and alleviate the symptoms of unfavorable movement of the rats’ left limbs.2.Acupuncture-rehabilitation Treatment can reduce the expression of Cx43 protein in the brain tissue of rats with cerebral ischemia,and up-regulate the expression of P-Cx43 protein and PKC protein in the brain tissue of rats with cerebral ischemia,inhibit the function of gap junction communication and accelerate the repair of brain tissue after ischemic injury.
