Objective:Observation method of needle kang game focal cerebral ischemia rat neural function defect scale and the influence of ischemic cortex RhoA,PTEN expressionMethods:Using Zea Longa method improved line switch method, making permanent focal cerebral ischemia model in rats.The rats were divided into 5 groups at random:model group,control group,acupuncture group and rehabilitation group,needle group,each group of 18 rats,according to the 3 d,7d and 14d three time points is divided into three subgroups,six in each group.24h after implementation of the corresponding intervention treatment,model group and control group without any intervention,clumps of head acupuncture point method acupuncture group, rehabilitation treatment group take the treadmill training,needle kang group applies the law to intervene.Each group in the corresponding time points by using improved neural function defect scale (mNSS) to assess the extent of neurologic deficits,immune histochemical methods are used to observe each group RhoA ischemic cortex of rats and the expression of PTEN positive cell number.Results:1. Improvement of nerve function defect score (mNSS):control group at each time point of neural function defect scale all without exception; Compared with the control group,model group at all time points of nerve function defect score were higher,the difference was statistically significant (P<0.05); After 7d,14d two time points and compared with model group,acupuncture group and rehabilitation group and neural function defect scale drop needle kang group were obviously,statistically significant difference (P<0.05).Compared with acupuncture and rehabilitation group,the needle mNSS score lower kang group rats is more apparent,difference has statistical significance (P<0.05).2. The expression of ischemic cortex RhoA:comparing with control group, model group all time points RhoA considerably increase the expression of positive cells,significant difference (P<0.05);After 3d,7d and 14d,compared with the model group,acupuncture group,the rehabilitation group and the needle kang group of RhoA protein expression was significantly decreased,statistically significant difference (P<0.05).Compared with acupuncture and rehabilitation group, needle kang group of RhoA reducing the most significant positive cells expression, had statistical significance (P< 0.05).3. The expression of PTEN protein in ischemic cortex:compared with control group,model group various points in the expression of PTEN increased significantly,the difference was statistically significant (P< 0.05);After 3 d,7 d and 14 d,compared with model group,acupuncture group,the rehabilitation group and the expression level of PTEN protein in needle kang group decreased significantly,had statistical significance (P< 0.05). Comparing with acupuncture and rehabilitation group,the needle were positive cells expression of PTEN level fell by the most prominent,the differences were statistically significant (P< 0.05).Conclusion:1. Needle kang method can obviously decrease cerebral ischemia rats nerve function defect score (mNSS),reduce the degree of nerve function defect, is better than the pure acupuncture and pure rehabilitation training.2. Needle kang method can cut in ischemic cortex RhoA and high expression of PTEN positive cells,may be a needle method is one of the mechanism of action of brain protection.
