| Objective:Observe the effect of Compound Qinglongyi Capsule on proteinuria level in the Adriamycin Nephropathy(AN)rats,and explore its therapeutic effect on kidney disease and the protective effect on podocytes from the perspectives of biochemistry and molecular biology,in order to offer new ideas of treating glomerular proteinuria clinically.Method:72 healthy and pure male SD rats were selected,12 were randomly selected as the blank group.The remaining 60 rats were given a one-time tail vein injection of adriamycin 6.5 mg/kg to establish adriamycin nephropathy(AN)rat models.The rats after successful modeling were randomly divided into 4 groups : Compound Qinglongyi Capsule group(Qinglongyi group),Tripterygium wilfordii polyglycoside tablets group(Tripterysium Glycosides group),and Benazepril hydrochloride tablets group(Benazepril group).Treat the rats after dividing them into groups,the Qinglongyi group was treated with a dose of 378mg/kg/d,the Tripterysium Glycosides group was treated with a dose of 9.45mg/kg/d,and the Benazepril group was treated with a dose of 1.05mg/kg/d,each group receiving the gavage at the dose of 2 ml once a day,while the control group and the model group receiving the gavage with the same amount of distilled water each day,which lasts 4 weeks.24-hour urine was collected by the end of the 1st,2nd,and 4th week to test the 24h-UPr of the rats and observe the general state of the rats.By the end of the4 th week,the rats were anesthetized and their blood was taken from the heart to detect Scr,BUN,TC,ALB,ALT and AST.Then the rats were killed and their kidneys were dissected.The average surface density ratio of CD2AP and PI3 K proteins in renal tissue was detected by immunohistochemistry,the expression of CD2AP mRNA and PI3 K mRNA in renal tissue was detected by real-time PCR,and the pathological changes of the kidneys were observed by transmission electron microscope.Results:After 4-week treatment,the general state of each group treated by medicine(Qinglongyi group,Tripterysium Glycosides group,Benazepril group)is better than before;24 hour urinary protein quantity decreased significantly(P<0.05),and the effect of Qinglongyi group and Tripterygium Glycosides group were similar,significantly stronger than that of Benazepril group(P<0.05);the value of TC all decreased,and the Qinglongyi group was the most significant(P<0.05);the value of ALB all increased,and the Qinglongyi group was the most significant(P<0.05);the expressions of CD2AP,PI3K protein and mRNA in podocytes were all significantly increased(P<0.05),and the results of Qinglongyi group is similar to the Tripterygium Glycosides group,which is significantly higher than that of the Benazepril group(P<0.05);the changes in the podocyte structure under the electron microscope are ameliorated,and the degree of foot process disappearance and fusion of glomerular podocytes in Qinglongyi group and Tripterygium Glycosides group was much more significantly reduced.Conclusion:Compound Qinglongyi Capsule can effectively reduce 24 hour urinary protein quantity in AIN rats with,improve their general state,decrease the content of serum total cholesterol and increase the content of serum albumin,raise the expression of CD2AP,PI3K protein and mRNA in podocytes,and reduce kidney pathological damage,slow the process of kidney disease. |
