Effect Of Compound Qinglongyi Capsule On Proteinuria And Oxidative Stress In Rats With Adriamycin Nephropathy

Objective: In this study,animal experiments were conducted to observe the effects of Compound Qinglongyi Capsule on 24-hour urinary protein quantity,serum Scr,BUN,TC,ALB,ALT and AST,the expression of SOD,GSH-Px,MDA in renal tissue in adriamycin nephrotic rats,and on renal pathological morphology.From the point of view of oxidative stress,it provides a theoretical basis for Compound Qinglongyi Capsule in the treatment of adriamycin nephrotic rats' proteinuria and the application of antineoplastic Chinese patent medicine in the treatment of renal disease.Methods: 72 6-week-old male SD rats were randomly divided into 2 groups according to their body weight:a control group(n = 12)and a model group(n = 60).According to the Bertani method,the rats in the model group were injected with adriamycin(doxorubicin)solution into the tail vein according to the dose of 6.5mg/kg body weight to establish the model.After successful modeling,the successful rats were randomly divided into four groups: Compound Qinglongyi Capsule group(Qinglongyi group),Tripterygium wilfordii polyglycoside tablets group(Tripterysium Glycosides group),Benazepril hydrochloride tablets group(Lotensin group)and model control group(model group).The general state of rats was observed every day.At the end of 4 weeks after the successful establishment of the model,the 24h-UPr of rats was measured.At the end of the 4th week,the rats were injected intraperitoneally with 10% chloral hydrate solution.After anesthesia,blood was taken from the heart to detect Scr,BUN,TC,ALB,ALT and AST.Taking the kidney,some of them were prepared into 10% tissue homogenate under the condition of ice water bath,and the content of SOD,MDA and GSH-Px in rat kidney were determined by xanthine oxidase method,thiobarbituric acid method and chemical colorimetry,respectively,and the other part was used to observe the pathological changes of kidney by electron microscope.Results: 1.The 24h-UPr of rats in the Qinglongyi group,the Tripterysium Glycosides group and the Lotensin group was lower than that in the model group(P<0.05),but there was no significant difference in 24h-UPr between the Qinglongyi group and the Tripterysium Glycosides group(P>0.05),and 24h-UPr in the Qinglongyi group was significantly lower than that in the Lotensin group(P<0.05);2.The value of TC in theQinglongyi group,the Tripterysium Glycosides group and the Lotensin group was lower than that in the model group,especially in the Qinglongyi group(P<0.05),and the value of TC in the Qinglongyi group was significantly lower than that in the Tripterysium Glycosides group and the Lotensin group(P<0.05).The value of ALB in the Qinglongyi group,the Tripterysium Glycosides group and the Lotensin group was higher than that in the model group,especially in the Qinglongyi group(P<0.05),and the value of ALB in the Qinglongyi group was significantly higher than that in the Tripterysium Glycosides group and the Lotensin group(P<0.05);3.The activities of SOD and GSH-Px in renal tissue of rats in the Qinglongyi group,the Tripterysium Glycosides group and the Lotensin group were higher than those in the model group,while the content of MDA was lower than that in the model group(P<0.05),but there was no significant difference in the activity of SOD,GSH-Px and the content of MDA in renal tissue between the Qinglongyi group,the Tripterysium Glycosides group and the Lotensin group(P>0.05);4.The results of transmission electron microscope showed that the degree of disappearance of foot process fusion of glomerular podocytes in Qinglongyi group and Tripterygium Glycosides group was significantly reduced.Conclusion:1.Compound Qinglongyi Capsule can reduce the 24-hour urinary protein level in rats with adriamycin nephropathy,thus delaying the progression of kidney disease;2.Compound Qinglongyi Capsule can increase the content of serum albumin and decrease the content of serum total cholesterol in rats with adriamycin nephropathy;3.Compound Qinglongyi Capsule can reduce the renal pathological damage in rats with adriamycin nephropathy,so as to play a role in renal protection;4.Compound Qinglongyi Capsule may reduce the damage of oxidative stress and reduce proteinuria by increasing the activity of SOD and GSH-Px and reducing the content of MDA.
