| Objective: The expression levels of RNA binding protein(CUGBP1)in serum and peripheral blood white blood cells of the four groups of subjects(physical examination certificate of health,chronic viral hepatitis B,post-hepatitis cirrhosis and hepatitis B-related primary liver cancer(PHC))were detected to explore the correlation between CUGBP1 and PHC.Method:(1)From August 2018 to August 2019,32 cases of healthy subjects(HE group),36 cases of hepatitis B group(CHB group),39 cases of cirrhosis group(HC group)and 43 cases of liver cancer group(PHC group)were enrolled in the four groups.Enzyme linked immunosorbent assay(ELISA)was used to measure the expression of CUGBP1 in serum samples and to investigate the correlation between CUGBP1 and PHC.(2)Include 12 patients in each of the HE group,HC group,CHB group and PHC group;Quantitative real-time PCR was used to detect the relative expression of genes in Peripheral white blood cells(PWBC)of CUGBP1,and an in-depth discussion on the characteristics of gene expression of CUGBP1 in PBMC samples.Results:(1)Serum CUGBP1 was expressed in HE group,HC group,CHB group and PHC group,and the expression level in HC group,CHB group and PHC group was lower than that in HE group.There was statistical significance in different stages of chronic liver disease(HC group,CHB group,PHC group)compared with HE group(P < 0.001).The ROC curve of PHC group was drawn.When the Max Youden index(MYI)was 0.272,the AUC was 0.6285,the sensitivity(SE)was 72.1%,and the specificity(SP)was 60.7%.There was no significant difference in serum CUGBP1 among different stages of chronic liver disease(HC group,CHB group and PHC group)(P > 0.05).(2)In this study,13 patients in the PHC group were AFP negative.The expression of CUGBP1 in serum of these patients was detected and the ROC curve was drawn.AUC=0.6281 was not significantly different from that of the HCC group,which indicated that the serum CUGBP1 also has certain diagnostic value in AFP negative patients.(3)The combined detection sensitivity of serum CUGBP1,AFP,CEA and CA199 was 95.3%,which were better than the results of each index alone.(4)The relative expression level of CUGBP1 m RNA in Peripheral white blood cells of PHC group was lower than that in HE group,and the difference was statistically significant(P<0.05).There was no statistically significant difference in the expression level of CUGBP1 m RNA among HE group,HC group and CHB group.Conclusion:(1)Compared with the HE group,the expression levels of CUGBP1 in the serum and CUGBP1 m RNA in Peripheral white blood cells of the PHC group were down-regulated,which proved the correlation between CUGBP1 and PHC.(2)CUGBP1 has potential diagnostic value in AFP-negative PHC patients.(3)Combined detection of serum CUGBP1,AFP,CEA and CA199 can improve the diagnostic sensitivity of primary hepatocellular carcinoma and reduce the rate of missed diagnosis. |
