| Abnormal ovulation is the most common and complex t ype of ovulation disorders that s eriousl y affect women’s reproductive health.Among them,the most common is pol yc ystic ovary s yndrome(PCOS),with an incidence of about 6% to 20%;ovulation disorders have complex etiologies,which may be caused by genetic variation or interactions of env ironmental factors,but the pathogenesis is still unclear.Follistatin(FST)is a single-chain gl ycos ylated pol ypeptide isolated from follicular fluid.It has a certain regulatory effect on the promotion of oocyte maturation and earl y embryonic development.The results of previous animal experiments all support the upregulation of FST gene expression.It leads to obstructed follicle development,atretic follicle formation and infertilit y,which are very similar to some t ypical phenot ypes of PCOS.Adeno-associated virus(AAV)is an ideal vector.Compared with retroviruses,lentiviruses,and adenoviruses,it has many advantages,such as non-pathogenicit y,wide infection range,and long-term In vivo expression.Therefore,in this experiment,cynomolgus monkeys,which are very similar to human reproductive models,were selected as animal models,and the cynomolgus monkey FST gene overexpression AAV vector was successfull y constructed.First,the ovaries were injected in situ and the relevant phenot ypes of the exp erimental animals were observed for a long time.Changes,such as changes in menstrual cycle,ovulation time,and follicle size.Secondl y,we al so studied the expression of FST onl y on special cell subsets during mouse embryonic periods,which preliminaril y verified that FST may be a pathogenic factor of PCOS,and It provides support for the subsequent exploration of the causal relationship between abnormal ovulation and abnormal expression of FST gene,and also provides a reasonable reference for earl y det ection of some abnormal ovulation diseases in clinical practice. |
