Objective: To observe the curative effect of Zhu Liangchun Gout Decoction on intermittent gout.The mechanism of Zhu Liangchun Gout Decoction in the treatment of gout was discussed based on network pharmacology.Methods: 1.In this study,60 patients with intermittent gout were enrolled in a non-double-blind,stratified randomized trial.The patients were divided into three groups:Western medicine group,Chinese medicine group and Chinese and Western medicine group,with 20 cases in each group.Western medicine group to the cloth he oral treatment,Chinese medicine oral treatment group to liangchunzhu gout soup,soup combined Chinese and western medicine group to liangchunzhu gout the cloth he oral treatment,treatment of 4 weeks and 8weeks after in the detection of uric acid level,and compare the clinical efficacy of before and after treatment,symptom integral and a decrease in the level of uric acid,and observe each group therapy drug adverse reaction.2.TCMSP database was used to screen the active ingredients and corresponding targets of Zhu Liangchun Gout Decoction.Gene Card,TTD,OMIM and DRUGBANK database were used to collect the action targets of gout,and drug-disease co-action targets were screened out by We Shengxin software.Subsequently,the protein-protein interaction network was constructed through the String database,and the PPI network was constructed through Cytoscape 3.6.0software.Topological parameter analysis was carried out to screen out the important targets.Then Metascape database was used to perform GO biofunctional annotation and KEGG pathway enrichment analysis on the selected common targets.Results: 1.Three groups of drug treatment of intermittent gout can reduce the symptom score,the total effective rate is more than 75%;And can reduce the blood uric acid level.In reducing the level of blood uric acid,Zhu Liangchun gout decoction alone is not as good as febuxat.And Zhu Liangchun gout soup combined with febuxostat can play a synergistic role.In terms of adverse reactions,the incidence of abnormal liver function was 15% in the western medicine group and 10% in the Chinese and western medicine group,while there was no obvious adverse reaction in the traditional Chinese medicine group.2.It was predicted that 189 effective compounds and 76 co-acting targets of Zhu Liangchun Gout Decoction in the treatment of gout were found.After screening,TNF,IL1 β,IL6,IL4,CXCL8,STAT3,JUN,CCl2,Rela,IL10,IL2,IFNG,ICAM1,MAPK14,IL1 A and other core targets were obtained.These targets may be the action targets of Zhu Liangchun gout decoction.31 GO biological processes,48 GO molecular functions and 88 cell components were obtained by GO biofunctional annotation,which suggested that Zhu Liangchun gout decoction acted on different targets through a variety of ways.KEGG pathway enrichment analysis showed that AGE-RAGE signaling pathway,IL-17 signaling pathway,HIF-1 signaling pathway,NF-κ B signaling pathway were significant pathways,which might be the action pathway of Zhu Liangchun gout decoction.Conclusion: 1.Zhu Liangchun gout decoction can improve the clinical symptoms of patients with intermittent gout and reduce the level of blood uric acid without obvious adverse reactions.When Zhuliangchun gout decoction was used alone,the level of uric acid was lower than febuxostat.Combined with febuxostat in the treatment of gout can synergically improve the clinical efficacy of gout patients.2.Zhu Liangchun Gout Decoction may improve clinical symptoms of gout and reduce blood uric acid level through biological processes such as controlling inflammatory factors,regulating immunity,affecting cell differentiation,and affecting lipid metabolism.
