Study On Expression And Biological Functional Of LncRNA-IRAIN In Mantle Cell Lymphoma

Objective: To investigate the relative expression of lncRNA-IRAIN,and its biological functions in mantle cell lymphoma(MCL).It aimed to clarify pathogenesis of lncRNA-IRAIN in the occurrence and development of MCL,and provide new biomarkers and therapeutic targets.Methods: 1.The relative expression of lncRNA-IRAIN and LSD1 m RNA in MCL cell lines(Jeko-1 and Granta-519)was detected by qPCR.The correlation between lncRNA-IRAIN and LSD1 in MCL cells was studied by linear fitting.2.MCL cells were transfected with lentivirus over-expressing lncRNA-IRAIN.The expression of lncRNA-IRAIN was detected by qPCR.Cell proliferation was studied by CCK8 method.And the cell apoptosis and cell cycle were measured by flow cytometry.3.The expression of LSD1 m RNA in MCL cells after transfected with lncRNA-IRAIN was detected by qPCR.Western blot was used to detect the protein of LSD1 and cleaved caspase3 in MCL cells.Result:1.The expression of lncRNA-IRAIN was lower(Jeko-1: p=0.0002,Granta-519: p<0.0001)and LSD1 m RNA was higher(Jeko-1: p<0.0001,Granta-519:p<0.0001)in MCL cell lines than that in normal lyphocytes.The expression of LSD1 m RNA was negatively correlated with lncRNA-IRAIN(r=-0.9789,p=0.0007).2.After infected by lentivirus lncRNA-IRAIN,the expression of lncRNA-IRAIN in MCL cells was up-regulated(Jeko-1: p<0.0001;Granta-519: p=0.0003).Compared to the control,up-regulation of IRAIN inhibited cellls proliferation within 5 days,the cells growth was decreased with time,p<0.01.It induced cells apoptosis(Jeko-1:p<0.0001,Granta-519: p<0.001)and blocked cells cycle in G1 phase(Jeko-1:p<0.001,Granta-519: p<0.001).3.Up-regulation of lncRNA-IRAIN decreased LSD1 m RNA(Jeko-1: p<0.01;Granta-519:p<0.05)and protein of LSD1(Jeko-1: p<0.001;Granta-519: p<0.005)in MCL cells.And it improved the apoptosis-related protein cleaved caspase3(Jeko-1: p<0.01;Granta-519: p<0.005).Conclusions:1.Lnc RNA-IRAIN is down-regulation,and LSD1 m RNA is up-regulation in MCL cell lines.There is a negative correlation between the lncRNA-IRAIN and LSD1 m RNA.2.Up-regulating lncRNA-IRAIN inhibits the cells proliferation,induces cells apoptosis,arrests the cells in G1 phase,and block the process of cells mitosis.Lnc RNA-IRAIN might be a tumor suppressor gene,which involves in the occurrence and development of MCL.3.Up-regulating lncRNA-IRAIN could reduce the expression of LSD1 m RNA and its protein and cleaved apoptotic caspase 3.
