Objective To investigate the effects of different concentrations of PTEN-LONG protein on non-small cell lung cancer cells(A549)and human lung cancer cells(H1299).Methods The PTEN-Long DNA sequence was obtained from Gen Bank database and primers were designed.RNA extracted from HEK293 T cells was used as template to amplify PTEN-Long gene.Pc DNA3.1-PTEN-L eukaryotic recombinant plasmid was constructed and transfected into 293 T cells.PTEN-Long protein was purified.The apoptosis of non-small cell lung cancer cells(A549)and human lung cancer cells(H1299)with different concentrations of PTEN-Long protein(10,50,100 n M)was detected by flow cytometry and TUNEL staining.The effects of PTEN-Long protein with different concentrations on the expression level of related molecules in PI3K/AKT signaling pathway were detected by Western blot.Results1.After the eukaryotic recombinant plasmid pcDNA3.1-PTEN-L constructed in this experiment was transfected into HEK293 T cells,PTEN-Long protein was expressed successfully.The purified PTEN-Long protein was obtained by affinity chromatography.2.Flow cytometry and TUNEL staining method showed that different concentrations of PTEN-Long protein treatment groups in A549 and H1299 cell lines can promote non-small cell lung cancer cell apoptosis(P<0.05),and the higher the concentration,the apoptosis The higher the ratio.3.Western blot assay showed that PTEN-Long protein could significantly decrease the ratio of p-PI3K/PI3 K and pAKT/AKT in non-small cell lung cancer cells(P<0.05).Conclusion PTEN-Long protein can promote apoptosis of non-small cell lung cancer cells in a dose-dependent manner through PI3K/AKT signal pathway in vitro.
